Transdermal Delivery of Drugs via Microneedles

Diltiazem hydrochloride (DH), perindopril erbumine (PE) and 0.1M isotonic Phosphate Buffer Saline (PBS) were bought from Sigma Aldrich (St. Louis, MO, USA). Distilled de-ionized water was obtained from NanoPure Infinity Ultrapure water purification system (Barnstead, Dubuque, IA, USA). Stainless steel microneedle rollers (0.5mm) were bought from Pearl Enterprises LLC (Lakewood, New Jersey, USA). Electric hair clipper was purchased from Walgreens, a local pharmacy store (Vallejo, CA). Electric dermatome was purchased from Nouvag® (Goldach, Switzerland). Franz diffusion system (PermeGear, Hellertown, PA, USA) was used for experiments. Frozen porcine ears were purchased from Pel-Freez Arkansas LLC (Rogers, Arkansas, USA). DH and PE were reconstituted in only PBS prior to experiments.

Cardiac Contractile Performance Analysis

The cardiac contractile performance was evaluated by studying isolated papillary muscles from the LV as previously described [27 (link),64 (link)]. The following mechanical parameters were measured during isometric contraction: maximum developed tension (DT; g/mm2), maximum rate of tension development (+dT/dt; g/mm2/s), and decline (−dT/dt; g/mm2/s). Regulator mechanisms of Ca2+ influx and L-type calcium channel activity were analyzed by Ca2+ concentration extracellular elevation maneuver (response percentage) and elevation of extracellular Ca2+ concentrations (0.5, 1.5, 2.5, and 3.5 mM) in the presence and absence of diltiazem (10−5 M; Diltiazem Hydrochloride, Sigma® Aldrich, St Louis, MO, USA), a specific blocker of L-type calcium channels. A post-rest contraction maneuver (response percentage) and elevation of extracellular Ca2+ concentrations (0.5, 1.5, 2.5, and 3.5 mM) in the presence and absence of cyclopiazonic acid (CPA, 30 mM; Penicillium cyclopium, Sigma® Aldrich, St Louis, MO, USA), a highly specific blocker of SERCA2a, were performed to assess the potential of SERCA2a function. All variables were normalized per cross-sectional area of the papillary muscle (CSA). Seven papillary muscles with CSA >1.5 mm2 were excluded from analysis as they can present central core hypoxia and impaired functional performance [64 (link),65 (link)].

Binding Interactions of AAG with Drugs

Native human AAG (#SLCD7253) and drug compounds (clindamycin phosphate, diltiazem hydrochloride and lidocaine) were purchased from Sigma–Aldrich (Darmstadt, Germany). Warfarin sodium salt was a gift from the Department of Medicinal Chemistry, Faculty of Pharmacy and Biochemistry, Zagreb, Croatia. All chemicals were used without further purification. Immobilized SialEXO® (derived from Akkermansia muciniphilla and expressed in E. Coli) was obtained from Smart Enzymes™ Genovis (Lund, Sweden), and HEPES buffer was obtained from VWR International (Leuven, Belgium). All other reagents were of analytical grade or better. All solutions were prepared immediately before the experiments in order to minimize contamination and/or impair the stability of solutions. The solutions for microcalorimetric titrations were prepared in 25 mM HEPES at pH 7.40 containing 150 mM NaCl (Kemika, Zagreb, Croatia), using doubly distilled water and 5 M NaOH (Kemika, Zagreb, Croatia) for adjusting the pH value. AAG and drug solutions were prepared by dissolving the appropriate amount of solid in the given solvent.

LC-MS Quantification of Cardiovascular Drugs

Heparin coated blood collection tubes were purchased from International Scientifique Supplies Ltd. (Bradford, UK). An 8 mm diameter punch was acquired from Maun Industries Ltd. (Nottingham, UK).
Following De Montfort University's Ethics Protocols, fresh blank blood was obtained from informed volunteers.
