Diverse Transgenic Mouse Models for Gastric Cancer Research

Cxcr4-EGFP mice were kindly provided by Richard J. Miller (Northwestern University Medical School, USA). LSL-KrasG12D and LSL-Trp53R172H mice were provided by Dr. Kenneth Olive (Columbia University, USA). Apcflox mice were obtained from the National Cancer Institute (NCI). Lgr5-DTR-GFP mice were provided by Genentech. Cdh1flox, R26-mTmG, R26-LacZ, R26-TdTomato, R26-Confetti, R26-EYFP, Cxcl12flox, Tie2-Cre, Id2-GFP, and Cag-CreERT2 mice were purchased from the Jackson Laboratory. Cre recombinase was activated by oral administration of TAM (1–5mg/0.2mL corn oil, as indicated). All animal studies and procedures were approved by the ethics committees at Columbia University and the Academy of Sciences of the Czech Republic. Human stomach tissue sections were obtained from DGC patients who underwent surgical resection or endoscopic submucosal dissection from 2001 to 2012 at Gifu University Hospital, Gifu, Japan. All study protocols were approved by the ethics committees, and written informed consent was obtained from all patients.

Genetic Mouse Models of Cancer

All mouse strains used have been previously reported: aP2-Cre (Tang et al., 2008 (link)), Adipoq-Cre (#10803, Jackson Laboratories) (Eguchi et al., 2011 (link)), Ucp1-Cre (#24670, Jackson Laboratories) (Kong et al., 2014 (link)), Fabp4-Cre (#5069, Jackson Laboratories) (He et al., 2003 (link)), Flk1::Cre (Kdr::Cre) (#018976, Jackson Laboratories) (Motoike et al., 2003 (link)), R26-mTmG (#7676, Jackson Laboratories) (Muzumdar et al., 2007 (link)), R26-tdTomato (#7914, Jackson Laboratories) (Madisen et al., 2010 (link)), R26-LacZ (#3474, Jackson Laboratories) (Soriano, 1999 (link)), SmoM2 (#5130, Jackson Laboratories) (Mao et al., 2006 (link)), LSL-KrasG12D (#8179, Jackson Laboratories) (Jackson et al., 2001 (link)) and Cdkn2AFlox (Aguirre et al., 2003 (link)). For Kaplan-Meier tumor free survival analysis, Adipoq-Cre;SmoM2/+ and Ucp1-Cre;SmoM2/+ animals were observed from birth and confirmed to be tumor free at necropsy. For Kaplan-Meier survival analysis, aP2-Cre; LSL-KrasG12D and aP2-Cre; LSL-KrasG12D;Cdkn2aFlox/Flox animals were observed from birth and sacrificed when showing signs of obvious tumor burden or other distress. Full necropsies were performed. All experimental procedures involving animals were reviewed and approved by SJCRH Institutional Animal Care and Use Committee.

Lineage Tracing of Mist1-expressing Cells

All mouse strains were maintained on a C57/Bl6 background. Mist1CreERT2 mice were produced as described (Shi et al., 2009 (link)) and crossed with the reporter strains R26LacZ (Gt(ROSA)26Sortm1Sor), R26Brainbow2.1 (Gt(ROSA)26Sortm1(CAG-Brainbow2.1)Cle/J), or R26TdTomato (Gt(ROSA)26Sortm9(CAG-tdTomato)Hze/J) (all from Jackson Labs). Standard genotyping was performed. All procedures and protocols were approved by the University Committee on Animal Resources at the University of Rochester.

LSL-KrasG12D and LSL-Trp53R172H mice were provided by Dr. Kenneth Olive (Columbia University). Cckbr-GFP BAC transgenic mice were purchased from MMRRC (GENSAT project [38 (link)]). Lgr5-DTR-GFP mice were provided by Genentech. Apcflox/flox mice were obtained from the National Cancer Institute. Lgr5-EGFP-IRES-CreERT, Sox2-EGFP, R26-mTmG, R26-TdTomato, R26-LacZ, Cxcl12flox/flox, and Tie2-Cre mice were purchased from the Jackson Laboratory. FucciG1 mice were obtained from RIKEN BRC. Cre recombinase was activated by oral administration of tamoxifen (TAM, Sigma, 3 mg/0.2 ml corn oil). All animal studies and procedures were approved by the ethics committees at Columbia University and the University of Tokyo. All mice were bred under specific pathogen free conditions. Comparisons were made with age- and sex- matched control animals.
