Examining ACE2KO Mice on High-Fat Diet

Ace2−/y mutant mice (ACE2KO) backcrossed into the C57BL/6 background for at least eight generations were used in the current study (18 (link),21 (link),22 (link)). All experiments were performed in accordance with University of Alberta institutional guidelines, which conform to guidelines published by the Canadian Council on Animal Care and the Guide for the Care and Use of Laboratory Animals published by the U.S. National Institutes of Health (revised 2011). Male wild-type (WT) and ACE2KO mice were fed either a high-fat diet (HFD) (45% kilocalories from fat) or a control diet (CON) (10% kilocalories from fat) from weaning to 6 months of age. ALZET microosmotic pumps (Model 1002; DURECT Corporation) were implanted subcutaneously in ACE2KO-HFD mice to deliver Ang 1-7 (24 μg/kg/h) or saline (control) for 4 weeks (21 (link)). All mice were studied at 6 months of age. Epicardial adipose tissues (EATs) were collected under a stereo microscope after removal of the pericardium and pericardial fat.

Angiotensin II-Induced Hypertension in GPER Mice

All procedures were approved by the Tulane University Institutional Animal Care and Use Committee and are in accordance with the NIH Guide for the Care and Use of Laboratory Animals. Heterozygous breeders30 ,31 (link) were used to create homozygous male and female wildtype (wt) and global G protein-coupled estrogen receptor (GPER) knockout (ko) mice that were maintained at Tulane University vivarium, and GPER expression was confirmed by genotyping. Mice were provided with water and food ad libitum in a temperature-controlled room (65-75°F) with a 12 h light to dark cycle. Alzet® micro-osmotic pumps model 1002 (DURECT Corporation, Cupertino, CA) containing Ang II (Bachem Americas Inc, Torrance, CA) were implanted subcutaneously to infuse at a rate of 700 ng/kg/min for two weeks. Mice infused with Ang II that did not reach a systolic blood pressure > 130 mmHg were excluded from the study. Mice were euthanized by isoflurane overdose and exsanguination at 20 ± 3 weeks of age, and carotid arteries were excised for biaxial pressure myography followed by constitutive modeling and histology.

Continuous Subcutaneous Insulin Delivery

Alzet Micro-osmotic pumps, model 1002 with pumping rate 0.25μl/hr (DURECT Corporation) were filled with Humulin (Eli Lilly) diluted in 1X sterile PBS to allow Insulin release of 0.2 or 0.3U/day. Pumps were implanted subcutaneously under isoflurane anesthesia. The mice were allowed to recover and their plasma glucose was monitored using glucose meter (ACCU-CHEK Aviva, Roche). Plasma insulin levels was measured by ELISA (Crystal Chem, Ultrasensitive mouse insulin ELISA kit, Catalogue #90080, Lot # 16SEUMI411 that detects both human and mouse insulin)
