Human embryonic kidney 293 (HEK293) cells were cultured in DMEM (Gibco, USA) supplemented with 10% heat-inactivated FBS, penicillin (100 U/mL), and streptomycin sulfate (100 μg/mL). The THP-1 human monocytic cell line was cultured in RPMI 1640 (Gibco, USA) supplemented with 10% heat-inactivated FBS, penicillin (100 U/mL), and streptomycin sulfate (100 μg/mL). All cells were maintained at 37°C with 5% CO2.

FMDV Isolation and Titration Protocol

The FMDVs (O/MYA98/BY/2010 and A/GD/MM/2013) were preserved and provided by OIE/National Foot-and-Mouth Disease Reference Laboratory of China. Swine kidney cells (IBRS-2) were purchased from CCTCC (China Center for Type Culture Collection) and then maintained in our laboratory. IBRS-2 cells were cultured in Dulbecco's Modified Eagle Medium (DMEM) (Gibco, Grand Island, NY) containing 10% fetal bovine serum (FBS) at 37 °C and 5% CO 2 . Escherichia coli (E. coli) Rosetta (DE3) Lys cells were used for expression of PoIFN-δ8. The TCID 50 of FMDV was calculated according to the Reed and Muench method [12] .
