Imaging and Quantification of Luciferase Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
In vivo imaging and quantification of luciferase activity were carried out using a cooled CCD camera (NightOwl II LB 983 NC-100; Berthold Technologies) and the provided software. Seeds from representative lines were simultaneously sown and grown on 0.6% agarose or MS agar plates in the presence or absence of 5 μM PAC or 5 μM PAC plus 5 μM GA. Plates were directly transferred to a chamber at a constant temperature of 22°C and a photoperiod of 16 h light/8 h dark for 24 or 48 h. Plates were then sprayed with 100 μM luciferin and imaged after 40 min.

Protein-Protein Interaction Imaging in Nicotiana

Check if the same lab product or an alternative is used in the 5 most similar protocols
Luciferase complementation imaging (LCI) for testing the interaction of PIF5 with COP1 and SPA1 in N. benthamiana was performed as previously described with few modifications (Chen, et al. 2008 (link)). A. tumefaciens strain GV3101 was used for the injection of different constructs into N. benthamiana leaves. 1mM Luciferin was sprayed into the N. benthamiana leaves and kept in the dark for 5 min. Low-light cooled CCD camera (NightOWL IILB 983 NC 100, BERTHOLD) was used to capture luciferase luminesence. An exposure time of 2 min with 2×2 binning was used. Relative luciferase was equivalent to luminescence intensity / 0.2mm2 leaf area.

Bioluminescence Imaging for Tumor Monitoring

Check if the same lab product or an alternative is used in the 5 most similar protocols
Bioluminescence imaging was conducted using bioluminescence technology (Night OWLII LB983 NC100, Berthold, Germany). Before imaging, all mice received 2.5mg of D-luciferin (Xenogen Corporation, Hopkinton, MA) intraperitoneally. Mice receiving the tail vein injections were imaged weekly in this same way to monitor the condition of tumor growth.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
