Quantifying Immune Markers in Breast Cancer

Check if the same lab product or an alternative is used in the 5 most similar protocols
CCL11 content of the serum of patients with BC and healthy individuals was measured by ELISA, according to the manufacturer's protocol. The Human Eotaxin ELISA kit (cat. no. ab185985; Abcam) was used to measure CCL11 levels, according to the manufacturer's protocol. The IL-2 and TGF-β1 content of the media of CD4+ T cells from the PBMCs of patients with BC or healthy individuals was measured using the Human IL-2 Quantikine ELISA kit (cat. no. S2050; R&D Systems) and the Human TGF-β1 Quantikine ELISA kit (cat. no. SB100B; R&D Systems), respectively. The CCL11, IL-2 and TGF-β1 content of the serum of tumor-bearing mice was measured using the Mouse CCL11/Eotaxin Quantikine ELISA kit (cat. no. MME00; R&D Systems), the Mouse IL-2 Quantikine ELISA kit (cat. no. SM2000; R&D Systems) and the Mouse/Rat/Porcine/Canine TGF-β1 Quantikine ELISA kit (cat. no. SMB100B; R&D Systems), respectively.

Cytokine Profiling of S-2P Stimulated Splenocytes

Check if the same lab product or an alternative is used in the 5 most similar protocols
Two weeks after the second injection, mice were euthanized and splenocytes were isolated and stimulated with S-2P protein (2 μg/well) as previously described26 (link). For detection of IFN-γ, IL-2, IL-4, and IL-5, the culture supernatant from the 96-well microplates was harvested to analyze the levels of cytokines by ELISA using Mouse IFN-γ Quantikine ELISA Kit, Mouse IL-2 Quantikine ELISA Kit, Mouse IL-4 Quantikine ELISA Kit, and Mouse IL-5 Quantikine ELISA Kit (R&D System). The OD450 values were read by Multiskan GO (Thermo Fisher Scientific).

I-A^b-Restricted Eα-Specific T-Cell Activation Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The I-Ab restricted Eα-specific 1H3.1 T-cell hybridoma cell line was used for activation assays to recognize preactivated dendritic cells expressing I-Ab::Eα52–68 complexes. T cells were cultured in RPMI-1640 medium supplemented with 10% heat-inactivated fetal bovine serum (Wisent) and 55 μM β-mercaptoethanol at 37°C with 5% CO2. For activation assays, preactivated DCs were mildly fixed in 1% PFA for 15 minutes at room temperature. Following fixation, cells were washed with PBS 3 times and then quenched thrice in complete medium for 10 minutes each at room temperature. After, 1H3.1 T cells and fixed DCs were co-cultured at 2:1 and incubated for 40 h at 37°C with 5% CO2. Next, the tissue culture medium was collected, and T cells were immediately isolated following centrifugation at 800g for 5 minutes. The supernatant was immediately stored in −80°C for downstream IL-2 secretion analysis. To quantify T-cell activation, secreted IL-2 samples were diluted 1:10 and subsequently analysed using Mouse IL-2 Quantikine ELISA Kit (R&D Systems Inc, Minneapolis, MN) as per manufacturer’s specifications.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
