Quantification of Bioactive Spice Compounds

All organic solvents were High‐performance liquid chromatography (HPLC) grade (Thermo Fisher Scientific, Fairlawn, N.J., U.S.A.). Deionized water was purified by the Milli‐Q system (Millipore, Bedford, Mass., U.S.A.). Reference standards apigenin, capsaicin, dihydrocapsaicin, cinnamic acid, cinnamaldehyde, p‐coumaric acid, bisdemethoxycurcumin, demethoxycurcumin, curcumin, luteolin, luteolin‐3‐glucuronide, piperine, rosmarinic acid, and vanillin were purchased from Sigma‐Aldrich (St. Louis, Mo., U.S.A.). Ferulic acid was purchased from USP (Rockville, Md., U.S.A.) and 6‐gingerol from Chromadex (Irvine, Calif., U.S.A.). They were used for the identification and quantification of spice chemicals.

Ginger Phytochemicals and Cell Viability

LPS from Escherichia coli O55:B5, adenosine 5’-triphosphate (ATP), phorbol 12-myristate 13-acetate (PMA), dimethyl sulfoxide (DMSO), and 3-(4,5-dimethyldiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) for cell culture experiment were acquired from Sigma-Aldrich (St. Louis, MO, USA). The ginger phytochemicals, including 6-gingerol (C17H26O4), 8-gingerol (C19H30O4), 10-gingerol (C21H34O4), 6-shogaol (C17H24O3), 8-shogaol (C19H28O3), and 10-shogaol (C21H32O3) with a purity of >99% were procured from ChromaDex (Irvine, CA, USA). Cell culture medium and reagents were obtained from Thermo Fisher Scientific (Grand Island, NE, USA). All other chemicals used were analytical grade.

(MACHEREY-NAGEL INC, PA, USA), iQ TM SYBR ® Green Supermix, and iScript ™ Reverse Transcription Supermix for RT-qPCR were purchased from Bio-Rad (Hercules, CA, USA). HO-1 antibody was purchased from Enzo Life Science (Farmingdale, USA). NRF-2 antibody was purchased from Abcam (Cambridge, UK).
