Confirming miRNA and sRNA Expression

Northern blot analysis was used to confirm the expression levels of known miRNAs and verify the new miRNAs and some other new sRNAs following the previously published protocol (Lopez-Gomollon et al. 2012). Briefly, 5 µg of total RNA from stem, root and leaf tissues were denatured and loaded into denaturing 16% polyacrylamide gel. The RNA was transferred to Hybond NX (Amersham) nylon membrane through semi-dry electrophoresis transfer system (Bio-Rad), and chemical cross-linking was done at 60°C for 90 minutes by using 1-ethyl-3-(dimethylaminopropyl) carbodiimide (Sigma). The probes were generated by labelling the oligonucleotides that were reverse complementary to the sRNAs of interest with γP32-ATP. The membranes were hybridized with the probes at 37°C overnight. The list of the probe sequences used for northern blot is provided in supplementary Table 1.

Trisodium citrate dehydrate and NaBH4 were purchased from Nanjing Chemical Reagent Co., Ltd. (Nanjing, China). CoCl2 was purchased from Guangdong Guanghua Sci-Tech Co., Ltd. (Guangdong, China). SARS-CoV-2 S protein RBD and mouse anti-human IgG (catalog no. V90401) were purchased from Nanjing Genscript Co., Ltd. (Nanjing, China). Monoclonal antibodies against the SARS-COV-2 RBD (GHMA 105–1 and GHMA 105–2) were purchased from Goodhere Biological Technology Co., Ltd. (Hangzhou, China). Polyethylene glycol 2 W (PEG, MW, 20000) was purchased from Sinopharm Chemical Reagent Co., Ltd. All chemicals were used without further purification.
Serum samples were obtained from Hôpital Militaire d’Instruction Med V Rabat and stored at −80 °C until use. This study was approved by the Ethics Committee of the Science and Technology Department of Huazhong University of Science and Technology (certificate no. S1029).

B16-F10 mouse melanoma cancer cells were
purchased from American Type Culture Collection (ATCC, Manassas, VA).
LIVE/DEAD cell imaging kits were purchased from Invitrogen (Carlsbad,
CA). Sodium alginate (Protanal LF20/40) of high molecular weight (≈250
kDa) was donated by FMC BioPolymers (Philadelphia, PA). Trypan Blue,
MES hydrate, adipic acid dihydrazide (AAD), 1-hydroxybenzotriazole
(HOBT), 1-ethyl-3-(dimethylaminopropyl) carbodiimide (EDC), iron (II,
III) oxide powder, sodium chloride, activated charcoal, irinotecan
hydrochloride, 5-fluorouracil, mitoxantrone, Dulbecco’s modified
Eagle’s medium (DMEM), fetal bovine serum (FBS), penicillin–streptomycin,
trypsin–EDTA, and alginate lyase were all purchased from Sigma-Aldrich
(St. Louis, MO). Sixteen-well xCELLigence e-plates were purchased
from ACEA Biosciences, Inc. (San Diego, CA).
