Optimized Topical CLT Formulation

CLT was purchased from Pol-Aura (Różanowo, Poland). Castor oil was supplied from Microfarm (Zabierzów, Poland). Rapeseed oil was purchased from Bunge Polska Sp. z o.o. (Kruszwica, Poland). In addition, 90% Lactic acid, Poloxamer 188 (Pluronic F-68) was purchased from Pol-Aura (Różanowo, Poland). Poloxamer 407 (Pluronic F-127), poly(ethylene glycol) 200 (PEG200), sorbitan monooleate (Span 80), and polyoxyethylenesorbitan monooleate (Tween 80) were purchased from Sigma-Aldrich (St. Louis, MO, USA or Steinheim, Germany). Propylene glycol was supplied by Firma Chempur (Piekary Śląskie, Poland).
Acetonitrile for HPLC (ACN) was purchased from S.Witko (Łódź, Poland). Acetone, methanol, and potassium phosphate monobasic (KH2PO4) were obtained from Chempur (Piekary Śląskie, Poland). Isopropanol was purchased from Pol-Aura (Różanowo, Poland). Milli-Q water (<0.05 µS/cm), used in batches manufacturing and all experiments, was self-produced from Hydrolab Ultra UV (Hydrolab Sp z o.o., Straszyn, Poland). All the chemicals and reagents used in this study were analytical grade.
Market reference, i.e., 1% CLT cream, MycoHydralin (Bayer Healthcare, Loos, France), was purchased in a local retail pharmacy in France.

Pharmacological Modulation of Aspirin Pathways

Aspirin (ASA), purchased as dl-lysine acetylsalicylate (Aspégic injectable, Sanofi Aventis), was dissolved in sterile deionized water and resuspended in drinking water at 30 mg/l (low) (37 (link)), 180 mg/l (medium) (38 (link)), or 625 mg/l (high) (8 (link)). SC-560 (Cayman Chemical) dissolved in DMSO (Sigma-Aldrich) was resuspended at 24 mg/l in drinking water supplemented with 0.2% (vol/vol) polyethylene glycol 200 (PEG200) and 0.01% (vol/vol) Tween-20 (both from Sigma-Aldrich) (77 (link)). NS-398 (Cayman Chemical) dissolved in DMSO was resuspended at 12 mg/l in drinking water supplemented with 0.9% wt/vol sodium chloride (Sigma-Aldrich) (77 (link)). Picotamide (PICO; Abcam) dissolved in 100% ethanol (Sigma-Aldrich) was resuspended in drinking water at 30 mg/l. U46619 (Cayman Chemical) was diluted in DMSO and delivered at 50 μg/kg through a 180-mg/l aspirin solution. Vapiprost (Vapiprost hydrochloride, Santa Cruz Biotechnology) dissolved in sterile water was resuspended in drinking water at 20 mg/l. All drinking water contained 2% wt/vol sucrose (Sigma-Aldrich). Clopidogrel [(±)-Clopidogrel (hydrochloride), Cayman Chemical] dissolved in 100% ethanol was delivered in saline at 10 mg/kg/d through i.p. injection (56 (link)). Eptifibatide (Sigma-Aldrich) was resuspended in sterile water and delivered in saline at 0.5 mg/kg/d through i.p. injection (78 (link)).

Topical Ophthalmic Formulation Development

Diclofenac sodium (DS) and fluorescein sodium were handled by Sigma-Aldrich (Budapest, Hungary). Isopropanol (IPA) was obtained from Avantor (Radnor, PA, USA). Hydroxypropyl methylcellulose (HPMC) was provided by Colorcon (Budapest, Hungary). Polyethylene glycol 200 (PEG 200) was purchased from Merck KGaA (Darmstadt, Germany). Phenoxyethanol and Caprylyl Glycol were from Biesterfeld GmbH (Hamburg, Germany). Polyoxyl castor oil was kindly supplied by BASF SE Chemtrade GmbH (Ludwigshafen, Germany). Gattefossé (Saint-Priest Cedex, France) provided Caprylocaproyl Polyoxyl-8 glycerides and CP Kelco A Huber Company (Atlanta, GA, USA) provided xanthan gum. Deionized and purified water was used (Milli-Q system, Millipore, Milford, MA, USA) during the research. The cellulose acetate filter (Porafil membrane filter, cellulose acetate, pore diameter: 0.45 μm) was acquired from Macherey-Nagel GmbH & Co. KG (Düren, Germany). Additionally, 70% sodium laureth sulfate (SLES) was provided by Kao Chemicals Europe S.L. (Barcelona, Spain).
