Quantification of APAP and Resveratrol in Zhishi

APAP and resveratrol (RES) were purchased from Shanghai Source Leaf Biological Technology Co., Ltd. HPLC-grade acetonitrile, methanol and formic acid were obtained from Dikma Technologies Inc. (Beijing, China). Deionized water (18.2 MΩ) was obtained from a Milli-Q water purification system (Millipore, Bedford, MA, USA). Assay design enzyme-linked immunosorbent assay kit (ELISA) was obtained from Nanjing Jiancheng Bioengineering Institute. All the reference standards were obtained from Chengdu Mansite Bio-technology Co., Ltd. The Zhishi sample was collected from Jiangxi province of China, and its species, local name, collection location, year of collection and growing environment are listed in the complementary materials. Its dried specimens were deposited at the Institute of Basic Theory, China Academy of Chinese Medical Sciences, Beijing, P. R. China.

Deionized water was purified from a Milli-Q academic ultra-pure water system (Millipore, Milford, MA, United States). Other chemical reagents were of analytical grade. All the solutions were filtered through a 0.22 μm filter membrane before UPLC analysis.

Quantitative Analysis of Fumonisins

HPLC-MS Analysis of Phytochemicals

The standards of columbianadin and columbianetin acetate were isolated and purified from APR extract by our laboratory, which were identified by H-NMR, IR, and HPLC-MS. The purity of all analytes were more than 98%. C18 (50 μm, 60 A), C18-N (50 μm, 60 A), AQ C18 (50 μm, 60 A), NH2 (40–60 μm, 60 A), COOH (50 μm, 60 A), Diol (50 μm, 60 A), Florisil (60–100 μm, 80 A), PS (40–60 μm, 70 A), PEP (40–60 μm, 70 A) and PEP-2 (40–60 μm, 60 A) were supplied from Agela technologies. HPLC-grade acetonitrile and methanol were both provided by Dikma Technologies Inc., United States. HPLC-grade formic acid was obtained from Tedia Company Inc. (Tedia, Fairfield, OH, United States). Other chemicals were of analytical reagent grade. Ultrapure water was supplied by a Milli-Q academic ultra-pure water system (Millipore, Milford, MA, United States). All the solutions were filtered through a 0.22 μm filter membrane before UPLC analysis.

Quantitative Analysis of Creatine

Cre (99.8% purity) and Cre-d3 (98% purity) was purchased from Sigma (St. Louis, MO, USA) and Toronto Research Chemicals (Toronto, Canada), respectively. Cre stock solution was prepared at a concentration of 14.57 mmol/L in water. The working solutions at various concentrations (4.4, 8.8, 17.7, 44.3, 88.5, 177.0, 442.5 and 885.0 μmol/L) were prepared by appropriately diluting the stock solution with 20% methanol. Isotope-labeled Cre-d3 was used as internal standard (IS) and prepared with 20% methanol at 132 μmol/L in working solution. All of these solutions were stored at -20°C until use.
