Conditional Deletion of Cox-2 in Myeloid Cells

Adult wild-type mice of both sexes of the C57BL/6J strain were obtained from Janvier labs. Mice with selective deletion of Cox-2 in myeloid cells were generated by crossing mice possessing loxP sites flanking exons 4–5 of the Ptgs2 gene [18 (link)] with mice expressing Cre recombinase under the LysM promoter [19 (link)]. The mice were housed under constant ambient temperature (21°C), on a 12 h/12 h light/dark cycle (lights on at 7 A.M.) with food and water available ad libitum. All animal experiments were approved by the Animal Care and Use Committee at Linköping University (#1854/2018).

Controlled Conditions for C57BL/6j Mice

The mice were housed in nest-enriched polysulfone cages, with polysulfone bottles. Offspring were mixed at weaning to avoid potential litter effects, with no more than one male per litter per cage, and were allowed to grow to 8 weeks of age.

Murine BBB Permeability Assay

Offspring were mixed at the weaning to avoid potential litter effects with no more than one male per litter per cage, and were allowed to grow to 8 weeks of age. For these experiments, 9 cohorts each comprising 9 to 32 animals distributed equally between the 4 treatment groups, were used. Ultimately, 32 animals from two cohorts were used for BBB permeability assays, 96 animals from five cohorts were used for the capillary-enriched fraction procedure and Western blot analysis, and then 68 animals from three cohorts were used for the immunocytochemistry study.
