Annexin 5 fitc and pi kit

Manufactured by Thermo Fisher Scientific
Sourced in United States

The Annexin V FITC and PI kit is a laboratory reagent designed to detect and quantify apoptosis in cells. It contains Annexin V, a protein that binds to phosphatidylserine, and propidium iodide, a DNA-binding dye. This kit allows for the identification of early and late apoptotic cells through flow cytometry analysis.

Annexin V-FITC and TMRE Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Phosphatidylserine externalization was detected using an Annexin V FITC and PI kit (Thermo Fisher, Cat. No. V13242) following the manufacturer’s instructions. Briefly, cells were washed with cold PBS and resuspended in Annexin V binding buffer and stained with Annexin V and PI at room temperature and then analyzed immediately. To measure change in mitochondria membrane potential cells were treated with 200nM of tetramethylrhodamine, ethyl ester (TMRE; Abcam, Cat. No. ab113852) for 15min at 37 °C. Annexin V and TMRE-positive cells were detected using the Becton-Dickinson LSR18 machine and analysed with FlowJo version 7 software module.

Apoptosis and Cell Cycle Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Apoptotic cells were detected using Annexin-V-FITC and PI kit (Thermo Fisher Scientific, Waltman, MA, USA) following the manufacturer's instructions, and presented as the percentage (%) of Annexin-V+PI+ cells.
To determine cell-cycle distribution, cells were collected, washed with PBS three times, and fixed in 70% ethanol overnight. Fixed cells were then stained using FxCycle™ PI/RNase Staining Solution (Thermo Fisher Scientific) according to the manufacturer's instructions. All FACS analysis was performed on FACSCalibur (BD Biosciences, San Jose, CA, USA).

Quantifying Apoptotic Cell Populations

Check if the same lab product or an alternative is used in the 5 most similar protocols
Apoptotic cells were detected using Annexin-V-FITC and PI kit (Thermo Fisher Scientific) following the manufacturer’s instructions. The percentage (%) of apoptotic cells was calculated as the  % sum of Annexin-V+PI (representing early apoptotic cells) and Annexin-V+PI+ cells (representing late apoptotic cells).

Annexin V FITC and PI Cytometry Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The Annexin V FITC and PI kit (Thermo Fisher, Cat. No. V13242) was used following the manufacturer’s instructions. Briefly, cells were washed by PBS, trypsinized, and suspended in annexin V binding buffer. Cells were stained with annexin V and PI for 15 min and then analyzed. Analysis was performed using FlowJo version 7 software module.
For cell cycle analysis, the cells were fixed, treated with bovine RNase, and then stained with propidium iodide (Sigma-Aldrich) for 15 minutes at room temperature. Samples were subjected to analysis using the Becton-Dickinson LSR18 machine and FlowJo version 7 software module.

Apoptosis and Cell Cycle Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells were treated with indicated doses of SCH772984 and phenformin for 48 hours. Analysis of apoptotic cells by annexin V staining (Annexin V FITC and PI kit, Thermo Scientific) was done according to manufacturer's instructions. Cell cycle analysis was performed after fixation in 70% ethanol and incubation with PBS containing 0.5 μg/mL RNaseA (Sigma-Aldrich) and 50 μg/mL propidium iodide (PI; Sigma-Aldrich, St. Louis, MO). Data was acquired on a Canto Flow Cytometer (Becton Dickinson, Franklin Lakes, NJ) and analyzed using FlowJo software (Treestar, Ashland, OR).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
