Dulbecco’s modified Eagle’s medium (DMEM) and trypsin were purchased from PAA Laboratories GmbH (Pasching, Austria). Fetal calf serum was purchased from Invitrogen. [3H]5-HT ([3H]5-hydroxytryptamine; [3H]serotonin; 28.3 μCi/mmol) and [3H]DA ([3H]dihydroxyphenylethylamine, [3H]dopamine; 46 μCi/mmol) were purchased from PerkinElmer, Boston, MA. [3H]1-Methyl-4-phenylpyridinium ([3H]MPP+; 85 μCi/mmol) was supplied by American Radiolabeled Chemicals (St. Louis, MO). Paroxetine was from Santa Cruz Biotechnology, mazindole, serotonin, levamisole, cocaine, aminorex, nisoxetine, D-amphetamine, and monensin were purchased from Sigma–Aldrich Co.

Radioligand Binding Assay for Monoamine Transporters

D‐Amphetamine (D‐Amph) was sourced from Tocris (UK), paroxetine from Santa Cruz (USA), p‐chloroamphetamine (pCA) and GBR12909 were from Sigma‐Aldrich (USA). Hellmanex was from Hellma France S.A.R.L. (Paris, France). Radiochemicals are [3H] 1‐methyl‐4‐phenylpyridinium ([3H]MPP+; 80–85 μCi × mmol−1) from American Radiolabeled Chemicals (St. Louis, MO, USA) and [3H]5‐HT (28.3 μCi x mmol−1) from PerkinElmer (Boston, MA, USA). Other chemicals and cell culture supplies were obtained from Sigma‐Aldrich (St. Louis, MO, USA), and cell culture dishes were from Sarstedt (Nuembrecht, Germany).

Synthesis and Characterization of N-Ethyl Cathinones

N-Ethyl cathinones were synthesized in a racemic form as hydrochloride
salts as described in Section 3.3. Solutions for injection were prepared
daily in an isotonic saline solution (0.9% NaCl, pH 7.4). Cocaine
and methamphetamine hydrochloride were generously provided by the
Spanish National Institute of Toxicology and Dr. Riera laboratory
from Parc Científic de Barcelona, respectively. α-PVP·HCl
was synthesized as described in ref (6 (link)). Cell culture media [Dulbecco’s modified
Eagle’s medium (DMEM) high-glucose] was purchased from Sigma-Aldrich.
[3H]1-Methyl-4-phenylpyridinium ([3H]MPP+) was supplied by American Radiolabeled Chemicals (St. Louis,
USA). [3H]5-HT, [3H]imipramine, and [3H]WIN35,428 were purchased from PerkinElmer Inc. (Boston, MA, USA).
The NGF was supplied by Upstate Biotechnology (Lake Placid, NY). The
CCK-8 cell counting kit was purchased from Vazyme Biotech Co., Ltd.
All other reagents were of analytical grade and purchased from several
commercial sources.

Synthetic Cathinone Characterization

Ring-substituted α-aminovalerophenone derivatives were synthetized in racemic form as hydrochloride salts as described in the Supplemental Material. Chemical purity and identification of the synthetic cathinones was assessed by thin-layer chromatography (TLC), proton and carbon nuclear magnetic resonance (1H NMR, 13C NMR), infrared spectroscopy (IR), and mass spectrometry (MS). Solutions for injection were prepared daily in isotonic saline solution (0.9% NaCl, pH 7.4). Cocaine was provided by the Spanish National Institute of Toxicology. [3H]1-Methyl-4-phenylpyridinium ([3H]MPP+) was supplied by American Radiolabeled Chemicals (St. Louis, United States). [3H]5-HT was purchased from Perkin Elmer, Inc. (Boston, MA, United States). All other reagents were of analytical grade and purchased from several commercial sources.
