Three different concentrations of santhopine or nicotine solution (low, 50 nmol g -1 soil; middle, 250 nmol g -1 soil; high, 1000 nmol g -1 soil) were applied every 3 days for five times, as described previously (13) . Sterile water was applied as a control. Tubes were incubated at room temperature in the dark. The concentrations of santhopine or nicotine solution were adjusted every time to keep the soil water content ratio at 30%. After incubation, total DNA was extracted using DNeasy PowerSoil Kit (Qiagen, Hilden, Germany) according to the manufacturer's protocol. Extracted DNA was quantified using Qubit Quantification Platform dsDNA HS Assay Kit (Invitrogen, Carlsbad, CA, USA) and stored at -80°C.
