Immunohistochemical Analysis of Mouse Spleen

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse spleens were embedded in optimal cutting temperature compound and frozen over dry ice. 10-µm sections were cut on a cryostat, mounted on Superfrost Excell slides (Thermo Fisher Scientific), and fixed in −20°C acetone for 20 min. After rehydration in staining buffer (PBS, 1% goat serum, 1% BSA, 0.1% Tween-20), slides were stained with B220-PE (RA3-6B2; BD Biosciences), CD3-APC (500A2; Life Technologies) and PNA-FITC (Vector Laboratories). Images were acquired and analyzed by using a Leica DM6000B microscope at 10× magnification, a Leica DFL300 FX camera, and Leica Application Suite Advanced Fluorescence software. Analysis was performed by using ImageJ software.

Immunofluorescent Staining and Morphometric Analysis of Mouse Kidneys

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse kidneys were frozen in OCT compound (Thermo Fisher Scientific). Five-micrometer sections were fixed in −20°C acetone for 20 min, rehydrated in staining buffer (PBS, 1% goat serum, 1% BSA, and 0.1% Tween 20), and stained with IgG/FITC (Sigma-Aldrich), IgG2c/FITC (SouthernBiotech), or C3/FITC (MP Biomedicals). Images were acquired using a Leica DM6000 B microscope, Leica DFL360 FX camera, and Leica Application Suite X software. For glomerular IC quantification, images were obtained using a constant exposure and scored from 0 to 3 by two independent observers blinded to genotype. Paraffin-embedded sections of kidney were stained with Jones silver methenamine. Images were acquired using a Leica DM6000 B microscope, Leica DFL300 FX camera, and Leica Application Suite X software. Image analyses of glomerular volume and cell counts were conducted using ImageJ.

Detecting Glomerular Immune Deposits

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse kidneys were embedded in OCT compound and snap frozen over dry ice. 5μm sections were cut using Cryostat, mounted on slides and fixed in acetone for 20 mins at -200C. After the fixation, the slides were incubated with blocking buffer (PBS, 1% goat serum, 0.1% Tween-20, 1% BSA). The slides were subsequently stained with anti-FITC IgG, IgG2c and C3 in blocking buffer, washed and coverslipped with aqueous mounting media. Images were acquired using Leica microscope, Leica DFL300 FX camera and Leica software LAS X. Location of glomerular immune complex deposits was scored by a pathologist (C.E.A) blinded to genotype.

Immunohistochemistry of Mouse Spleen and Kidney

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse spleens and kidneys were embedded in OCT compound and snap frozen over liquid nitrogen. 10 μm sections were cut on a cryostat, mounted on Superfrost plus slides, and fixed in −20°C acetone for 20 minutes. After rehydration in staining buffer (PBS, 1% goat serum, 1% BSA, 0.1% Tween-20), slides were stained with: B220-PE, CD4-FITC and GL7-APC (spleen); or, IgG-FITC, IgG2c-FITC, or C3-FITC (kidney). Images were acquired using a Leica DM6000B microscope, Leica DFL300 FX camera, and Leica Application Suite Advanced Fluorescence software. For glomerular immune-complex quantification, images were obtained using a constant exposure and scored from 0–3 by two independent observers blinded to genotype.

Check if the same lab product or an alternative is used in the 5 most similar protocols
Frozen sections (embedded in OCT) of murine spleen and kidney were fixed in acetone, rehydrated in staining buffer (PBS, 1% goat serum, 1% BSA, 0.1% Tween-20), and stained with: B220-PE (BD Biosciences), CD3-APC (Life Technologies) and PNA-FITC (Vector Labs) (spleen); or IgG2c-FITC (Southern Biotech), or C3-FITC (MP Biomedicals) (kidney). Images were acquired with a Leica DM6000B microscope, Leica DFL300 FX camera, and Leica Application Suite Advanced Fluorescence software. Glomerular immune complexes were quantified by two independent observers blinded to genotype.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
