Glucose Tolerance Test in Aged Rats

After a 16-hour fast, animals were injected with glucose (2g/kg ip) at the ages of 4, 8, and 13 weeks. Plasma samples were obtained for insulin measurements in a separate cohort of animals at earlier time points. Plasma was isolated and stored at -80°C until used for radioimmunoassays. Since 11-month-old rats on a HFD had a qualitative increase in abdominal adiposity, intravenous GTT were conducted under anesthesia to enhance reliability of glucose administration and decrease procedural stress for these animals. In the 11-month-old animals, the rats were fasted and anesthetized with isoflurane before tail vein injection of glucose (0.5g/kg).
