Peptide Characterization by Mass Spectrometry

The quality of all chemicals was of analytical grade, unless stated otherwise. Disodium hydrogen phosphate dihydrate, ethanol, formic acid (FA; 98–100%), hydrochloric acid (32%, w/w), pentane, 1-propanol, potassium dihydrogen phosphate, sodium chloride, tris(hydroxymethyl)-aminomethane (TRIS), and urea were purchased from Merck (Darmstadt, Germany). IDAM was from Applichem (Darmstadt, Germany). α-Chymotrypsin (from bovine pancreas, TLCK-treated, ≥40 U/mg protein), pepsin (from porcine gastric mucosa, 3200–4500 U/mg protein), TCEP, trifluoroacetic acid (TFA; 99%), and trypsin (from bovine pancreas, TPCK-treated, ≥10 000 BAEE U/mg protein) were obtained from Sigma-Aldrich (Steinheim, Germany), and deuterated methanol-d4 containing tetramethylsilane (TMS) was from Euriso-Top (Gif sur Yvette Cedex, France). The peptide LQLQPFPQPQLPYPQPQLPYPQPQPLPYPQPQPF (33-mer) and the isotopically labelled peptide LQLQP*FPQPQLPYPQPQLPYPQPQLPYPQ*PQ*P*F (*33-mer) with *P: L-[13C5][15N]-proline and *F: L-[13C9][15N]-phenylalanine, were purchased from Genscript (Hongkong, PR China) with a purity of >90%. Water for HPLC was purified using an Arium 611VF water purification system (Sartorius, Goettingen, Germany).

Enzymatic Hydrolysis of Wheat Proteins

Pepsin (from porcine mucosa, 10 FIP U/mg), trypsin (from bovine pancreas, TPCK treated, 10.000 BAEE U/mg protein), α-chymotrypsin (from bovine pancreas, TLCK-treated, ≥40 U/mg protein), and thermolysin (from Geobacillus stearothermophilus, 30–175 U/mg protein) were purchased from Sigma Aldrich (Darmstadt, Germany) and Merck (Darmstadt, Germany).

Preparation of Fluorinated Glass Slides

were provided by Menzel-Gläser, Germany. Tetraethoxysilane
(TEOS, reagent grade, 98%), Trichloro (1H, 1H, 2H, and 2H-perfluorooctyl)
silane (PFOTS, 97%), hexadecane (reagentPlus, 99%), and deuterium
oxide (D2O, 99.9 atom % D) were purchased from Aldrich.
Ammonia (28%), toluene, acetone, and ethanol absolute were provided
by VWR CHEMICALS. N-Hexane (95%) was purchased from
Fisher Chemical. Ultrapure water was produced by Sartorius Arium 611
VF Water purification System with a resistivity of 18.2 MΩ·cm.
PS colloids with a diameter of 338 nm and 1.43 μm were prepared
using surfactant-free emulsion polymerization57 (link) and purified by several cycles of centrifugation and redispersion
in ultrapure water. Small and large particles therefore shared the
same surface chemistry.

Enzymatic Analysis of Wheat Flour Carbohydrates

Trypsin (from bovine pancreas, TPCKtreated, ≥ 10,000 BAEE U/mg protein), sodium hydroxide solution (50-52%; eluent for IC) and anhydrous sodium acetate (≥ 99%) were from Sigma-Aldrich (Steinheim, Germany). α-Amylase assay reagent (4-nitrophenyl α-dmaltoheptaoside (blocked), plus excess α-glucosidase and glucoamylase) and β-amylase assay reagent (4-nitrophenyl β-d-maltotrioside, plus excess thermostable β-glucosidase) were purchased from Megazyme (Bray, Ireland). d-( +)-Glucose (≥ 99.5%), d-(-)-fructose (≥ 99%), sucrose (≥ 99.5%), maltose monohydrate (≥ 99%), maltotriose (≥ 90%) and maltoheptaose (≥ 70%) were from Sigma-Aldrich; maltotetraose (≥ 97%), maltopentaose (≥ 90%), maltohexaose (≥ 95%) and maltooctaose (≥ 80%) from Santa Cruz (Dallas, USA). Wheat flour type 550, according to the German flour classification system, was provided by Rosenmühle (Ergolding, Germany).
