Cytokine ELISA from Mouse Blood Lysates

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole blood was collected from anesthetized mice via cardiac puncture. Blood Cell lysates for enzyme-linked immunosorbent assay (ELISA) were collected via sonication of cells in lysis buffer supplemented with protease and phosphatase inhibitors. ELISAs were performed for CCL2 (R&D, DY479), CCL7 (Boster Bio, EK0683), CCL8 (R&D, DY790), CCL11 (R&D, MME00), CCL12 (R&D, MCC120) and CCL5 (R&D, DY478-05) on cell lysates and cell supernatants according to the manufacturer’s instructions.

Quantifying Secreted Cytokine Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
Secreted protein levels were examined by ELISA as previously described using commercially available kits (for murine CCL11, R&D, catalogue# MME00; for human CCL11, R&D, catalogue# DY320; for murine IL-1β, R&D, catalogue# MLB00C; for murine IL-6, R&D, catalogue# M6000B; for murine MCP-1, R&D, catalogue# MJE00B) according to vendor’s recommendations. Data were normalised by cell number.

Quantifying Secreted Eotaxin Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
Secreted Eotaxin levels were examined by ELISA as previously described using commercially available kits (for eotaxin/CCL11, R&D, catalog# MME00; for IL-4, R&D, catalog# M4000B; for IL-10, R&D, catalog#M1000B; for IL-12, R&D, catalog#M1270) according to vendor’s recommendations. For measuring eotaxin levels in the liver, cut a small slice of the tissue (~100 μg) and homogenize in ice-cold PBS with freshly added protease inhibitor cocktail (Sigma, P-8340). Centrifuge at 14000 g at 4 °C for 15 min. Remove and aliquot the supernatant for ELISA. Data were normalized by cell number (for supernatant collected from primary cell culture) or tissue weight (for liver homogenates).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
