Lactoferrin Modulates Lung Cancer Immune Response

Human lung epithelial adenocarcinoma cells (A549) were bought from the cell resource center, Shanghai Institutes for Biological Sciences, Chinese Academy of Sciences. We purchased RPMI-1640 medium (500 mL), fetal bovine serum (FBS, 500 mL), and penicillin (100 U/mL)-streptomycin (100 mg/L) solution (100×, 100 mL) from Gibco. Recombinant human lactoferrin (partially iron saturated), with purity greater than 90%, was purchased from Sigma-Aldrich. The miR-146a primers, as well as the related reagents, were purchased from Invitrogen. The primers of TLR4, MyD88, IRAK1, TRAF6, NFKB, TNFA, IL1B, and GAPDH were purchased from Sangon. Real-time PCR reagents (for extraction and quantification of mRNA and miRNA, respectively) were purchased from Takara. Primary antibodies against ACTB, TLR4, MyD88, IRAK1, TRAF6, NFκB, TNF-α, and IL-1β, as well as the corresponding secondary antibodies, were purchased from Santa Cruz Biotechnology. The LPS, Cell Counting Kit-8, and ELISA kits for the determination of TNF-α and IL-1β, and reagents for the western blot assay were purchased from Solarbio. The enhanced chemiluminescence kit was purchased from Thermo Fisher Scientific.

Targeting ILK in A549 Lung Cancer Cells

MTT (thiazole blue) was purchased from Beyotime Institute of Biotechnology (Haimen, Jiangsu, People’s Republic of China); lentivirus coated ILK shRNA (Henan Biotools Biological Technology Co., LTD, Henan, People’s Republic of China) was also purchased.

Gene Expression Analysis in Rat Sciatic Nerve

TRIzol reagent (Thermo Fisher Scientific, USA) was utilized to extract total RNA from rat sciatic nerve tissues. Then cDNA was synthesized by reverse transcription according to the instructions of the reverse transcription PCR kit (Takara, Japan), and the synthesized cDNA was tested for concentration and purity. The real-time PCR reagent (Takara, Japan) was then utilized, and the reaction system was: 95°C for 1 min; 35 cycles of 95°C for 40 s, 58°C for 40 s, and 72°C for 45 s; and 72°C for 10 min. Quantitative analysis was carried out using the 2−ΔΔCt method. The primer sequence used is listed in Table 1.
