LN428 and LN308 were previously established from human GBM samples [23 (link)]. The human lung cancer cell line A549 and human breast cancer cell line MDA-MB-231 were obtained from ATCC (Cat. # CCL-185 and HTB-26). All cell lines were cultured in DMEM medium (Corning, 10013CV) supplemented with 10% fetal bovine serum (sigma, F0926),1% penicillin and 1% streptomycin, at 37°C with 5% CO2 supplementation. Clinical grade ribociclib (LEE011) was obtained from Novartis, Inc. Carmustine (C0400), carboplatin (C2538), temozolomide (T2577), etoposide (E1383), irinotecan (I1406) were from Sigma-Aldrich; paclitaxel or taxol (S1150) and palbociclib (S1116) were from Selleck Chemicals.

Palbociclib (S1116) and Abemaciclib (S7158) were purchased from Selleck Chemicals (Houston, Texas, USA). Antibodies against HSP90 (H-114), cyclin D1 (A12), CDK6 (C-21), CDK4 (DCS-35), p16 (C-20), p21 (H164), cyclin E (HE12), c-Jun (G4) and c-Fos (E8) were from Santa Cruz Biotechnology; antibodies against cyclin D2 (D52F9) and p-RB (S795) were from Cell Signaling; antibody against SMARCA4 were from Bethyl Laboratories (A300-813A). Antibody against Rb (554136) was from BD Pharmingen. Cyclin D3 (ab28283) antibody was from Abcam. Antibody against SMARCA4 was used with 1:5000 dilution and all others with 1:1000 dilution. Antibodies for IHC and ChIP are listed in the corresponding method section below.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
