Modulation of Autoimmune Responses in ARE-Del Mice

Female heterozygous ARE-Del +/− mice were generated by crossing male homozygous ARE-Del −/− mice [29 (link),30 (link)] with female wild type B6 mice. Mice were housed in specific pathogen free conditions at the University of California, Davis vivarium. 12 weeks old female ARE--Del+/− were randomly divided into four experimental groups and intraperitoneally (IP) administrated with either phosphate buffered saline (PBS, controls), anti-BAFF alone, anti-CD20 alone or anti-BAFF in combination with anti-CD20 mAbs. Anti-BAFF (10F4, GlaxoSmithKline, GSK), a neutralizing recombinant hybridoma monoclonal antibody raised against a mouse BAFF protein produced in Armenian Hamster cell line (No Hamster material present). Anti-BAFF was administered at 100 mcg (5 mg/kg) three times a week, anti-CD20 (Ultra-LEAF purified anti-mouse CD20, BioLegend) was administered at 250 mcg (12.5 mg/kg) once every 4 weeks for a total of 12 weeks. Serum immunoglobulin (Ig) G, IgA, IgM, AMA levels and inflammatory cytokine levels were monitored every 4 weeks. All studies were conducted in accordance with the University of California, Davis and the GSK Policy on the Care, Welfare and Treatment of Laboratory Animals and were reviewed and approved by the Institutional Animal Care and Use Committee at GSK and the University of California Animal Care and Use Committee.

Investigating CD73 in 4T1 Breast Cancer

Four-week-old female Balb/c mice (Balb/cOlaHsd) were obtained from Envigo (Netherlands). Animals were maintained under controlled pathogen-free environmental conditions with a 12h light/dark cycle. Mice were inoculated with sh-NT and sh-CD73 4T1cells (2×104 cells in 100 μl PBS per mouse) orthotopically into 4th mammary fat pads (n = 10/group) and followed for 31 days. For the metastasis models, the mice were inoculated intravenously with sh-NT and sh-CD73 4T1 cells (5×104 cells in 100 μl PBS per mouse) into tail vein (n = 6/group) and followed for 20 days. In the B cell depletion model, 100 µM/animal Ultra-leaf purified anti-mouse CD20 (BioLegend, 152104) and control IgG antibody (BioLegend, 400671) were injected intravenously in the tail vain, once cells were inoculated and followed for 34 days. Animals were treated intraperitoneally each 4th day with the dose of 6 µg ZOL/animal. Body weights and tumor dimensions (35 (link)) were measured once a week. The animals were sacrificed when weight loss was ≥ 10% (data not shown).
