Isolation and Expansion of NK Cells

Spleens were isolated and single cell suspensions were prepared by pressing through 100 μm cell strainers. NK cells were isolated using DX5-labeled MACS® beads according to the manufacturer's protocol (Miltenyi Biotec) and expanded in RPMI1640 complete medium supplemented with 5,000 U/ mL recombinant human IL-2 (Proleukin®, Novartis) for 5–7 d. Purity of NK cells was assessed by flow cytometry and were routinely 85–95% CD3ϵNK1.1+ of total cells. Cultivated NK cells were further stimulated with 5 ng/ mL recombinant mouse IL-12 (R&D Systems) for the times indicated and cells were used for Western blot analysis and supernatants for ELISA. For the analysis of the expression of G zmB, Prf1 and miRNAs, cells were deprived from IL-2 for 4 h prior to cell lysis.

In Vitro Polarization of TH1 Cells

Endotoxin-Induced Inflammation in Mice

CD4+ T Cell Activation Assay

CD4+ Tn isolated from WT B6 and T-KO mice were stimulated with anti-CD3 (2μg/ml, Biolegend) and anti-CD28 (2μg/ml, Biolegend) Abs together with recombinant human IL-2 (10ng/ml, R&D Systems) and recombinant mouse IL-12 (5ng/ml, R&D Systems). On Day 7, cells were restimulated in 96-well plates with plate-bound anti CD3 Abs (1μg/ml, Biolegend) for 5 hours before collecting the supernatants. The concentration of IFN-γ and IL-4 were measured in triplicate by using recombinant mouse IFN-γ and IL-4 ELISA kits in accordance with the manufacturer's instruction (Biolegend).

IL-12 Stimulation of NK Cells

Differential T Cell Polarization

Naïve CD4+ CD62L+ T cells from the spleens of unprimed OT-II TCR transgenic mice were purified using the MACS cell sorting system (Miltenyi Biotec). T cells were polarized with 10 ng/mL recombinant mouse IL-12 (R&D Systems), 5 ng/ml mouse IFN-γ (R&D Systems), 20 U/mL human IL-2, and 5 µg/mL anti-IL-4 (BD Pharmingen) for Th1 cell differentiation, 5 ng/mL human TGF-β1 (PeproTech), 20 ng/mL mouse IL-6 (R&D Systems), 20 U/mL human IL-2, and anti-IL-4 and anti- IFN-γ (2 µg/mL; BD Pharmingen) for Th17 cell differentiation, as well as 5.0 ng/mL human TGF-β1 (PeproTech), 100 U/mL human IL-2, anti-IL-4, and anti- IFN-γ (5 µg/mL; BD Pharmingen) for induced Treg (iTreg) cell differentiation.
