Sod kit wst

Manufactured by Dojindo Laboratories
Sourced in Japan

The SOD kit-WST is a laboratory equipment product developed by Dojindo Laboratories. It is designed to measure the activity of superoxide dismutase (SOD), an important antioxidant enzyme, in biological samples. The kit utilizes a water-soluble tetrazolium salt (WST) as the detection reagent, which produces a colored formazan product upon reaction with superoxide radicals.

Oxidized LDL Stress Biomarker Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 75 µg/ml ox-LDL treatment for 48 h, HUVECs were harvested and lysed. Then the cell extract was utilized for the determination of malondialdehyde (MDA), lactate dehydrogenase (LDH), and superoxide dismutase (SOD) activity, as per the user’s guidebook of the MDA kit (Beyotime), LDH kit (Beyotime), and SOD kit-WST (Dojindo, Kumamoto, Japan).

ROS Detection and Antioxidant Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
To detect ROS production, HK-2 cells were harvested and incubated with 5 μM 2ʹ,7ʹ-dichlorodihydrofluorescein diacetate (DCF-DA; Sigma-Aldrich) for 1 h at 37°C, followed by determining the fluorescence intensity at the wavelength 488/525 nm. Then, ROS generation was quantified using FACS-Calibur (BD Biosciences). To measure SOD and LDH activity, the cells were collected, lysed, and then cell extract was obtained. Then, the examination of SOD and LDH activity in cell extract was performed with a SOD kit-WST (Dojindo, Kumamoto, Japan) and LDH kit (Beyotime) based on the manufacturer’s specification [28 ].

Quantifying Oxidative Stress in Kidney Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
For the measurement of reactive oxygen species (ROS) generation, SV40-MES13 cells were collected after treatment with HG or NG treatment for 24 h, followed by incubation with 10 nM Dichlorofluoresceindiacetate (DCFH-DA, Beyotime, Shanghai, China) for 30 min at 37°C. And then, the samples were assessed under a fluorescence microscope (485nm excitation, 530 nm emission), followed by quantification with a FACS-Calibur (BD Bioscience). For the detection of Lactate Dehydrogenase (LDH) activity and Superoxide Dismutase (SOD) activity, treated SV40-MES13 cells were harvested and lysed, followed by an examination in the cell extract using LDH kit (Beyotime) and a SOD kit-WST (Dojindo, Kumamoto, Japan), in line with the manufacturer's instructions.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
