Comprehensive Analysis of Cytokine Expression

Check if the same lab product or an alternative is used in the 5 most similar protocols
One million freshly isolated PBMCs were incubated overnight at 37°C, 5% CO2 in Iscove's Modified Dulbecco's Medium (GIBCO) supplemented with 10 % fetal calf serum (GIBCO) and 1% Penicillin/Streptomycin (GIBCO) in a 96-well plate. Cell were then treated with GolgiPlug (BD) to inhibit intracellular protein transport and stimulated with IL-2, PMA (0.05μg/ml) and Ionomycin (1μg/ml) for 4 hours at 37°C, 5% CO2. Subsequently, cells were stained for surface markers CD3 (SK7, BD), CD4 (SFCI12T4D11, Beckman Coulter) and CD45RO (UCHL1, BD) and intracellular markers IL17 (eBio64DEC17, eBioscience), IL-4 (8D4-8, eBioscience) and IFN-γ (B27, BD) and measured by flow cytometry. CD3+CD4+CD45RO+ cells were analyzed for cytokine expression levels.

Murine Erythroleukemia Cell Line FBL as a Model for Cancer Immunotherapy

Check if the same lab product or an alternative is used in the 5 most similar protocols
FBL is a murine erythroleukemia cell line of B6 origin that expresses the H-2b-restricted Friend murine leukemia virus-derived Gag glycoprotein as a tumor associated antigen [9] (link), [28] . Gag peptide (CCLCLTVFL) and ovalbumin (SIINFEKL) control peptides were purchased from Pi Proteomics. Cell culture was conducted in complete RPMI-1640 with 10% FBS (Sigma). Fluorochrome-conjugated antibody to mouse CD8 (53–6.7) was purchased from eBiosciences. Fluorochrome-conjugated mouse antibodies to CD90.1 (OX-7), IFNγ (XMG1.2), TNFα (MP6-XT22), and human antibodies to CD4 (RPA-T4), CD8 (RPA-T8), and CD45RO (UCHL1) were purchased from BD Biosciences. Fluorochrome conjugated antibodies to mouse neuropilin-1 (761705) and human neuropilin-1 (446921) were purchased from R&D Systems. efluor-670 cell dye was purchased from Invitrogen. Anti-mouse Nrp-1A blocking antibody was provided by Genentech and administered intraperitoneally at 10 mg/kg every other day, similar to studies previously described [45] (link).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
