Enzymatic Synthesis and Characterization

ω-pentadecalactone (PDL), diethyl sebacate (DES), N-methyldiethanolamine (MDEA), 3,9-Divinyl-2,4,8,10-tetraoxaspiro[5.5]undecane (DTSU), ethyl glycolate, ethyl 6-hydroxy hexanoate and methyl 10-hydroxydecanoate were purchased from Sigma Aldrich Chemical Co. and were used as received. Immobilized Candida antarctica lipase B (CALB) supported on acrylic resin (Novozym 435), potassium t-butoxide, chloroform, dichloromethane, hexane, pentane, diphenyl ether, tetrahydrofuran, ethylene diamine, triethylamine and chloroform-d were also obtained from Sigma Aldrich Chemical Co. The lipase catalyst was dried at 50 °C under 2.0 mmHg for 20 h prior to use. Plasmid DNA (pGL4.13) encoding the firefly luciferase (pLuc) and Luciferase Assay Buffer were obtained from Promega Co. (Madison, WI). GFP reporter gene pSicoR-GFP (pGFP) was obtained from Addgene.

The following reagents were used as received: N,N-dimethyl formamide (DMF), chloroform (CHCl3), ethyl acetate (EtOAc), n-hexane, sodium sulfate (Na2SO4), methanol (MeOH), methylene chloride (CH2Cl2), sodium bicarbonate (NaHCO3), sodium hydroxide (NaOH), N,N,N′,N′-tetramethyl-O-(benzotriazol-1-yl) uronium hexafluorophosphate (Kanto Chemical Co., Inc., Tokyo, Japan), N-ethyl-N-(1-methylethyl)propan-2-amine (Wako Co., Tokyo, Japan), stearylamine, silver (I) carbonate, N,N′-dicyclohexyl carbodiimide (DCC), 1-hydroxybenzotriazole (HOBt) (Kishida Chemical Co., Oshaka, Japan), di-tert-butyl-dicarbonate (TCI Co., Tokyo, Japan), hydrobromic acid (acetic acid solution) (Merck KGaA Co., Tokyo, Japan), α-d-cellobiose octaacetate, α-d-Glucose pentaacetate, ethyl 6-hydroxy hexanoate, dimethyl sulfoxide (DMSO), and deuterated solvents (CDCl3, DMSO-d6 and D2O) (Sigma-Aldrich, St. Louis, MO, USA). N,N-diisopropyl ethylamine (DIEA) were purchased from Wako Pure Chemical Industry Co., Inc. (Tokyo, Japan). Di-Boc-lysine (2) was synthesized according to the previous literature [24 (link)].
