Biodegradable PLA-PTMC Film with Oregano Oil

Commercial poly(lactic acid) (PLA, Mw = 280kDa, Mw/Mn = 1.98) from Natureworks LLC (Blair, NE, USA) was used as the biodegradable matrix. Poly(trimethylene carbonate) (PTMC, Mw = 100 kDa, Mw/Mn = 1.70) was synthesized in our laboratory. According to previous reports, [13 (link),14 (link)] using trimethylene carbonate as monomer and 1.5‰ stannous octoate as catalyst, polytrimethylene carbonate (PTMC) was synthesized by ring-opening polymerization under the 130 °C for 4 h. Oregano essential oil was supplied by dōTERRA Co., Ltd. (New York, NY, USA). Chloroform was obtained from Chengdu Kelong Chemical Co., Ltd. (Sichuan, China). Folin-Ciocalteu reagent, sodium carbonate, gallic acid standard and 2,2-diphenyl-1-picrylhydrazyl (DPPH) were purchased from Sigma Chemical Co. (St. Louis, MO, USA). All other reagents used in this study were of analytical grade.

Synthesis of SEBS Copolymer Membranes

Tri-block copolymer SEBS YH-501, with number-average molecular weight of 60 000 g mol−1, was supplied by Sinopec (Yue Yang). 1,4-Bis(chloromethoxy)butane (BCMB) was synthesized according to literature.24 (link) Pyrrole was obtained from Aladdin and distilled under nitrogen atmosphere before use. Chloroform, xylene, stannic chloride, dimethyl sulfoxide (DMSO), sodium hydrogen carbonate, potassium iodide, lactate, benzaldehyde, ethanol and methanol were all of analytical grade and used as received from Chengdu Kelong Chemical Reagent (Chengdu).

Fabrication of PLA/AgNP Composites

Silver nanoparticles (AgNPs) were purchased from Wanjing New Material Co., Ltd. (Hangzhou, China), with an average particle size < 150 nm and purity of 99.0%. Tributyl acetocitrate (ATBC), a plasticizer, and chloroform, a solvent, were supplied by Chengdu Kelong Chemical Co., Ltd. (Sichuan, China). The reagents used in this work were of analytical grade.

Cytotoxic Evaluation of XY-4 in Melanoma

N-[1-(2,3-dioleoyloxy) propyl]-N,N,N-trimethylammonium methyl sulfate (DOTAP) and cholesterol were purchased from Avanti Polar Lipids (Alabaster, AL, USA). DMEM and MTT were obtained from Sigma-Aldrich Co., (St Louis, MO, USA) and used without further purification. Chloroform and other organic solvents were purchased from ChengDu KeLong Chemical Co., Ltd (ChengDu, People’s Republic of China) Hoechst 33258 was purchased from Beyotime, (Shanghai, People’s Republic of China). XY-4 was previously synthesized in our laboratory. Mouse B16 melanoma cell line was purchased from the American Type Culture Collection (Manassas, VA, USA). Cells were cultured in DMEM medium supplemented with 10% fetal calf serum, incubated at 37°C with 5% CO2 humidified air atmosphere. Antibodies against mouse cytochrome c, β-actin, Bcl-xl, caspase 3 and caspase 9 were purchased from Abcam (Cambridge, MA, USA). C57 mice were obtained from Beijing HFK Biotechnology Co., Ltd (Beijing, People’s Republic of China) and maintained under specific pathogen-free conditions. All animal procedures were approved and controlled by the Institutional Animal Care and Treatment Committee of Sichuan Provincial People’s Hospital and carried out according to the Animal Care and Use Guidelines of Sichuan Provincial People’s Hospital.

Graphene-Reinforced Polymer Microspheres

Divinylbenzene (DVB), sodium persulfate (Na2S2O8) and sodium dodecyl sulfonate (SDS) were obtained from Aldrich, Shanghai, China. Graphene (GN, lamellae size 5–15 µm) was supplied by Deyang Carbonene Technology Co., Ltd., and the lamellae were composed of five to six layers of graphene. Absolute ethanol, petroleum ether, chloroform, toluene and acetone were all purchased from Chengdu Kelong Chemical Reagent Factory. Ultrapure deionized water from an ultrapure water machine was used to prepare all solutions.
