GC-MS Analysis of Amino Acids

During purification, an internal amino acid standard was added to samples, which was a mixture of three non-biological amino acids: nor-valine, nor-leucine, and 4-chloro-phenylalanine. The method described by Wanek et al. (2010 (link)) was developed further at ISOFYS, Ghent University (Andresen et al. 2015 (link)). Concentrations of amino acids were determined using gas chromatography–mass spectrometry (GC–MS, Trace GC–DSQ, Thermo Fisher). Separation was done on a VF 5-MS column (30 m × 0.25 mm ID × 0.25 µm film). With the available technical capacity, we focused on 14 amino acids: alanine, glycine, valine, leucine, serine, isoleucine, threonine, proline, asparagine, aspartate, methionine, glutamate, phenylalanine, and tyrosine.

Determination of Soil Amino Acids

The method to assess the individual free amino acid (FAA) content and isotopic enrichment is described in detail elsewhere (Wanek et al. 2010; Andresen et al. 2015 Andresen et al. , 2016a)) . In short, at arrival at ISOFYS lab, the SPE columns were washed with 10 mL of ultrapure water after which the FAAs were eluted with 30 mL 3 MolÁL -1 NH 4 OH. The NH 4 OH extraction solvent was removed by evaporation at reduced pressure (c. 30 mbar by rotovap), and amino acids were derivatized by ethyl chloroformate in ethanol/pyridine to block the polar groups of amino acid in order to make them volatile for GC seperation. Finally, the individual FAAs were measured by gas chromatographymass-spectrometry (GC-MS, Trace GC-DSQ, Thermo Fisher). Detection limit for individual amino acids is lower than 0.02 mgÁkg -1 soil.
