Human CRC HCT-8 cells and 5-FU resistant HCT-8/5-FU cells were obtained from Nanjing KeyGen Biotech. Co., Ltd. Cells were grown in RPMI-1640 medium, supplemented with 10% FBS, 100 U/ml penicillin and 100 µg/ml streptomycin. In addition, HCT-8/5-FU cell cultures were supplemented with 15 µg/ml 5-FU. Cells were cultured at 37°C and 5% CO2 in a humidified incubator. The cells were subcultured at a ratio of 1:4-1:6 at between 80 and 90% confluency.
