Apoptosis and Cell Cycle Analysis of Cervical Cancer Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Flow cytometer analysis was further used to detect the apoptotic cervical cancer cells. Briefly, the cells were exposed to RTF for 24 h and subsequently stained with FITC conjugated Annexin V/PI, and the fluorescence signals were analyzed by a flow cytometer (B75442, Beckman, Pasadena, CA, USA). The percentage of cells in early apoptosis was calculated by Annexin V-positivity and PI-negativity, while the percentage of late apoptotic cells was calculated by Annexin V-positivity and PI-positivity.
In addition, flow cytometry was also performed to analyze the cell cycle arrest of cervical cancer cells induced by RTF treatment for 24 h and subsequently stained with propidium iodide (PI).

Aggregation Analysis of K562 Cells and Nuclei

Check if the same lab product or an alternative is used in the 5 most similar protocols
K562 cells were collected by centrifugation (300 g, 5 min, room temperature). 5 × 105 cells were used for cell aggregation analysis, and 5 × 105 cells were used to extract nuclei for nucleus aggregation analysis. The labeling procedures for cells and nuclei were like before. For cells and nuclei, 5 and 2.5 µl assembled CASB complex was used, respectively. After labeling, DAPI was added to nuclei suspension according to manufacturer’s instruction. Afterward, cells and nuclei were imaged by fluorescence microscopy (Eclipse Ts2‐FL, Nikon) and analyzed by flow cytometry (B75442, Beckman Coulter).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
