Synthesis and Characterization of WCN-21 Compound

WCN-21 (purity > 95%) was synthesized in our lab. Dimethyl sulfoxide (DMSO) and methyl alcohol were purchased from Sinopharm Chemical Reagent Co., Ltd (Shanghai, China). 3-(4, 5-dimethyl-thiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) was purchased from Sigma-Aldrich Chemical Co. (St Louis, MO, USA). All solvents and reagents were of analytical or HPLC grade and used without further purification.
Dulbecco’s modified eagle medium (DMEM) was purchased from Sigma-Aldrich (St. Louis, MO, USA). Fetal bovine serum (FBS) was purchased from Zhejiang Tianhang Biological Technology (Hangzhou, China). M-plasmocin was purchased from Invivogene (San Diego, CA, USA). Cell lines HepG2, Hep3B, Huh7, and L02 were obtained from the China Center for Type Culture Collection at Wuhan University (Wuhan, China).

Recombinant Protein Expression in E. coli

The expression vector pET28a(+) and E. coli strains BL21 (DE3), were stored at − 80 °C in our lab. Kanamycin, protease inhibitors phenylmethanesulfonyl fluoride (PMSF) and IPTG were purchased from Sangon Biotech (Shanghai, China). p-NPG was purchased from Bio Teke Corporation (Beijing, China). Glycine, Tris, SDS, Bromophenol blue, coomassie brilliant blue R 250 and p-nitrophenol (p-NP) were purchased from San gon Biotech (Shanghai, China). Acrylamide and TEMED were bought from Sigma (USA). Glycerinum, isopropanol, methyl alcohol, β-mercaptoethanol, ammonium persulfate and glacial acetic acid were obtained from Sinopharm Chemical Reagent (Shanghai, China). Thermo Scientific Pierce BCA Protein Assay Kit was purchased from Thermo Fisher Scientific (MA, USA). High Affinity Ni-NTA Resin was bought from Genscript (Nanjing, China). All other reagents were of analytical grade and used without any further treatment.

Clonogenic Assay for Radiation Sensitivity

A total of 2×105 log-phase 5637 cells were seeded into 35 mm culture dishes (Thermo Fisher Scientific, Inc.) and subjected to increasing doses of γ-rays (0, 2 and 5 Gy). At 4 h post-irradiation, the cells were diluted serially to appropriate concentrations (100, 200 and 800 cells per 3 ml) and seeded into 60 mm dishes in triplicate. Following 14 days of incubation at 37°C, the colonies were rinsed twice with phosphate-buffered saline (PBS; Beyotime Institute of Biotechnology, Haimen, China), fixed for 15 min using methyl alcohol (Sinopharm Chemical Reagent Co. Ltd., Shanghai, China), stained with 0.1% crystal violet solution (Sangon Biotech Co., Ltd., Shanghai, China). The visible colonies (>50 cells) were counted and the surviving fraction was calculated.

Collagenase, Celastrol (CSL) and interleukin 1β (IL-1β) were obtained from Sigma-Aldrich, USA. Ethylenediaminetetraacetic acid (EDTA), 10% paraformaldehyde, horseradish peroxidase (HRP)-conjugated secondary antibody, Cell Counting Kit-8 (CCK-8), RIPA lysis buffer, BCA Protein Assay Kit and enhanced chemiluminescence (ECL) detection kit were obtained from Beyotime Institute of Biotechnology, China.

For cytotoxicity tests, human embryo hepatocytes (L-02 cells) were purchased from Procell (Procell Co. Ltd., Wuhan, China). Culture medium RPMI-1640 and fetal calf serum were purchased from HyClone ((GE-Healthcare, Logan, UT, USA). The 0.05% trypsin solution was from Thermo Fisher (Thermo Fisher Scientific, Waltham, MA, USA).
