Cell Culture Protocols for Drug Screening

Caki-2 cells were cultured in McCoy’s 5A (modified) medium supplemented with 10% FCS, Calu-3 cells were cultured in MEM with 10% FCS and 1x non-essential amino acids and both obtained from ATCC (Manassas, VA, USA). HEK293T cells were cultured in DMEM supplemented with GlutaMAX, 10% heat-inactivated FCS. Caco-2 cells were from Sigma Aldrich (Schnelldorf, Germany) and were cultured in EMEM medium supplemented with 10% FCS, L-glutamine and non-essential amino acids (M7145, Sigma Aldrich, Schnelldorf, Germany). HepG2 was obtained from Sigma Aldrich (Schnelldorf, Germany) and cultured in DMEM supplemented with GlutaMAX and 10% heat-inactivated FCS. All media contained 1% penicillin/streptomycin and the cells were cultured at 37 °C in a 5% CO2 atmosphere. Silvestrol was dissolved in DMSO and further diluted in media (cstock = 6 mM, maximal DMSO concentration during experiments 0.1% v/v). EGTA, famotidine and carbamazepine were from Sigma Aldrich (Schnellendorf, Germany). Lucifer Yellow (sc-215269) was obtained from Santa Cruz. Celecoxib was synthesized by WITEGA Laboratorien Berlin-Adlershof GmbH (Berlin, Germany). Forskolin and ionomycin were obtained from Sigma Aldrich (Schnelldorf, Germany). Silvestrol was provided by the Sarawak Biodiversity Centre (Kuching, North-Borneo, Malaysia; purity > 99%) and zotatifin was bought from MedChemExpress (USA, purity: 98%).

ETEC Challenge Model in Mice

Groups of eight 6-week-old female BALB/c mice (Harlan Sprague Dawley) were immunized s. c. with 50 μL of sterile PBS (pH 7.4, Fisher Scientific) or formulations containing aglycosylated PD or PD-O148 conjugate, according to the 49-day immunization schedule described above. At 7 days post-vaccination (48 h prior to challenge infection), mice received streptomycin (5 g/L) and fructose (6.7% (w/v)) in the drinking water to eradicate normal flora and fecal pellets were collected. Food was withheld 12 h prior to challenge infection and replaced with sterile water without antibiotics. Famotidine (50 mg/kg) (Sigma-Aldrich) was then administered 2 h prior to challenge infection to neutralize gastric acid. At 9 days post-vaccination, mice were subsequently infected with a 200-μL inoculum containing ∼1 × 104 CFU of ETEC strain B7A, administered by gavage with a feeding needle directly introduced in the stomach via the esophagus. After challenge infection, mice were checked daily for 3 days, and fecal pellets were collected and stored at −20°C for further analysis. Mice were sacrificed 72 h post-infection. All procedures were carried out in accordance with protocol 2012-0132 approved by the Cornell University Institutional Animal Care and Use Committee.

Monitoring Gastric Acid Secretion in hGOs

Acid secretion assays were performed as previously described (Schumacher et al., 2015). hGOs were grown in the chambered coverglass (Thermo Scientific) and the chamber was placed on an inverted confocal microscope (Zeiss LSM 710), and experiments were performed under 5% CO2 and 37°C conditions (incubation chamber, PeCon, Erbach, Germany).
Freshly isolated mouse gastric fundic glands or cultured hGO were incubated with acridine orange (10 μM), then acridine orange fluorescence was excited at 458 nm or 488 nm and images were collected at 600–650 nm (Red) or 500–550 nm (Green), respectively. On the other hand, to monitor hGOs luminal pH, the ratiometric pH sensitive dye, 5-(and-6)-carboxy SNARF-5F (5mM stock: EX 560 nm, EM 565–605 (Green) and 620–680 (Red) nm: Invitrogen) was microinjected (46–92 nl) into the lumen and monitored. Fluorescent dye also added into medium. Histamine (100 μM; Sigma) was added to media, while famotidine (100 μM; Sigma) or omeprazole (100 μM; Sigma) were pre-incubated at least 30 min before Histamine. Images were analyzed using MetaMorph software (Molecular Devices, Downingtown, PA). Background corrected 620-680/565-605 nm ratio values were converted to pH using a standard curve.
