The strain of EV71 CC077 was a gift from Professor Baisong Zheng of Jilin University, Changchun. The viruses were propagated in African green monkey kidney cells (Vero). Vero (No CCL81) and human rhabdomyosarcoma RD (No CCL136) cells were purchased from the ATCC (Manassas, VA, USA) and were maintained in Dulbecco’s modified Eagle’s medium (DMEM) (Hyclone, Logan, UT, USA) supplemented with 10% fetal bovine serum (FBS) (GIBCO BRL, Grand Island, NY, USA).
Quantification of Ginsenosides by iEESI-MS
Three ginsenoside standards samples, namely, ginsenoside Rb1, ginsenoside Re, and ginsenoside Ro, were provided by Shanghai Yuanye Biotechnology Co., Ltd. (Shanghai, China, HPLC purity > 98%). Deionized water (18.2 MΩ/cm) was prepared using a Milli-Q Water Purification System (Billerica, MA, USA). HPLC-grade methanol was purchased from ROE Scientific Inc. (Newark, DE, USA). AR grade NH4Cl and NH4Ac were purchased from Xilong Chemical Co., Ltd. (Xilong, Guangxi, China). HPLC grade C2H5OH, HCOOH, and CH3CN were purchased from Anpel Laboratory Technologies (Shanghai) Inc. (Shanghai, China).
Ginsenoside standard samples were dissolved in methanol to obtain standard stock solvents of all three target analytes at a final concentration of 100 μg/mL. The solvents were stored at 4 °C prior to the iEESI-MS analysis.
Quantification of Ginsenoside Compounds
Acetonitrile was purchased from Fisher Scientific (Waltham, MA, USA). Water was purified through a Milli-Q water purification system (Millipore, MA, USA). TNF-α, IL-1β, IL-6 and NO ELISA kits were provided by Jiangsu Mai Sha Industry Co., ltd (Yancheng, China). Dextran sulfate sodium (DSS) was obtained from MP Biomedicals (California, USA). Cell Counting Kit-8 (CCK-8) was obtained from Biosharp Life Science (Shanghai, China). Lipopolysaccaride (LPS) was purchased from Sigma-Aldrich (St. Louis, MO, USA). Griess reagents were purchased from Promega (Madison, USA).
