Serum Metabolite Profiling by LC/MS

For LC/MS analysis, 30 μL of patient serum was mixed with 470 μL of 85% methanol in water containing the internal standards (12.5 ng/mL kynurenine-d6, 100 ng/mL tryptophan-d5, and 12.5 ng/mL serotonin-d4). After removing proteins via centrifugation (18,440 or 21,210 ×g, 10 min, 4 °C), the solution was filtered with FastRemoverC18 (GL Science, Tokyo, Japan) using an automated system (Microlab NIMBUS with an MPE2 unit; Hamilton, Reno, NV, USA). The flow-through fraction was diluted 1:3 (v/v) with water and subjected to LC/MS analysis. The preparation method used for the serum samples of the healthy subjects was slightly different because of the low volumes of patient serum samples available for analysis. Proteins were removed using an automated system by mixing the serum (50 μL) with 450 μL of 90% methanol in water containing the internal standard and filtering with FastRemover for Protein (GL Science); the following procedure was the same as described above.

Protein and Lipid Removal for IC-MS

Check if the same lab product or an alternative is used in the 5 most similar protocols
Extraction was performed using an automated system (Microlab NIMBUS, Hamilton Robotics, Reno, NV). The sample (20 µl) was mixed with 80 µl water and 400 µl methanol containing internal standard (2 µM hippuric acid-d5, Toronto Research Chemicals). Subsequently, 300 µl of the mixture was loaded onto the FastRemover for Protein (GL Sciences, Tokyo, Japan) to filter out protein coagulants. The flow-through solution (200 µl) was subsequently loaded onto the FastRemover C18 (GL Sciences) to remove lipids. Finally, 100 µl of flow-through solution was completely evaporated and stored at -80 °C until required for IC-MS analysis.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
