Synthesis and Procurement of Pharmacological Agents

FPFT-2216 and pomalidomide were synthesized by Fujimoto Pharmaceutical Group. Avadomide, iberdomide, siremadlin, idasanutlin, navtemadlin, milademetan, and MG-132 were procured from MedChemExpress; Z-VRPR-FMK and phorbol 12-myristate 13-acetate (PMA) were procured from Adipogen Life Sciences. Lenalidomide and ionomycin were obtained from Cayman Chemical, BAY 11-7082 from FUJIFILM Wako Pure Chemical Corporation, and safimaltib from Selleck Chemicals. Rituxan Intravenous Infusion (rituximab) was obtained from Zenyaku Kogyo. Z-VRPR-FMK was dissolved in PBS, and other reagents were dissolved in DMSO. The final DMSO concentration used for in vitro experiments did not exceed 0.25%.

Cell Viability Assay with CCK8

Cell viability was detected by CCK8 assay (C6030, New Cell & Molecular Biotech Co., Ltd). Briefly, cells were seeded in 96-well plates with a density of 2 × 103 cells/well. After treatments, CCK8 was added to the cell culture medium and maintained for additional 1 h. The absorbance was determined at 450 nm by a microplate reader (BioTek). Rhabdoid Tumor and Rhabdomyosarcoma Cell Culture

The human malignant rhabdoid tumor cell line JMU-RTK-2 was obtained from the JCRB cell bank and cultured in Dulbecco’s modified Eagle’s medium supplemented with 5% fetal bovine serum. The rhabdomyosarcoma cell line RMS-YM was obtained from RIKEN BRC cell bank and cultured in RPMI-1640 supplemented with 10% fetal bovine serum, 20 mM-HEPES, and 0.1 mM None-essential amino acids (FUJIFILM Wako Pure Chemical Corporation). All the cells were maintained at 37 °C in a humidified atmosphere containing 5% CO2. Topoisomerase inhibitors, etoposide, doxorubicin, and epirubicin were purchased from FUJIFILM Wako Pure Chemical Corporation (Osaka, Japan), ellipticine was purchased from Merck (DS, Darmstadt, Germany), topotecan hydrochloride hydrate was purchased from Sigma Aldrich (St Louis, MO, USA), and siremadlin was purchased from Medchemexpress (Monmouth Junction, NJ, USA).

Siremadlin and Trametinib Combination in Melanoma

PEG 400 (catalogue number 81172) and Cremophor RH40 (catalogue number 07076) were provided by Merck (formerly Sigma-Aldrich), EtOH (catalogue number 1016/12/19) was provided by POCH, and Labrafil M1944CS (catalogue number 178290) was provided by Gattefosse.
