Hypoxia-Inducible Factor Protein Analysis

EnVision™ kits, horseradish-peroxidase (HRP)-conjugated goat anti-mouse immunoglobulin G (IgG) and goat anti-rabbit IgG were obtained from Dako/Agilent (Santa Clara, CA, USA). A polyclonal anti-HIF-1α antibody was from Cayman (Ann Arbor, MI, USA). Anti-HIF-2 and anti-HIF-1β antibodies were obtained from Novus Biologicals (Litteton, CO, USA), and anti-LDH-A antibody was from Proteintech (Rosemont, IL, USA). Dulbecco’s modified Eagle’s medium (DMEM) with pyruvate and IFN-γ were obtained from Wako Pure Chemical Industries Ltd (Osaka, Japan) and DMEM without pyruvate was obtained from Nacalai Tesque (Kyoto, Japan). Fetal bovine serum (FBS) was from Equitech Bio (Kerrville, TX, USA). An anti-β-actin antibody and oxamate were purchased from Sigma (St Louis, MO, USA). Isogen was obtained from Nippon Gene (Toyama, Japan) and the ReverTra Ace qPCR RT Kit was from Toyobo (Osaka, Japan). The Ambion PARIS™ Kit and TaqMan gene expression master mix were purchased from Applied Biosystems (Carlsbad, CA, USA). HIF-1α Chromatin Immunoprecipitation Assay

ChIP assay was conducted using a ChIP assay kit (Upstate, Lake Placid, NY, USA) as previously described [8]. Protein samples were crosslinked using 1 M MDSP (Pierce Biotechnology, Waltham, MA, USA). Cells were lysed in low-stringency IP buffer containing protease inhibitor cocktail (Roche) and endonuclease (Pierce Biotechnology). The antibodies were coated with A/G magnetic beads (Pierce Biotechnology). HIF-1α was immunoprecipitated with anti-HIF-1α antibody (Cayman, Ann Arbor, MI, USA). Anti-rabbit IgG antibody (Millipore) was used as a control. The immunoprecipitated complex was washed with the lysis buffer, eluted in SDS-PAGE buffer containing 50 mM DTT, and boiled for 10 min, followed by SDS-PAGE separation under reducing conditions. After transferring onto a PVDF membrane (Perkin-Elmer), the proteins were incubated with the anti-HIF-1α (Cayman) or anti-c-Myc (Abcam, Cambridge, UK) antibody overnight at 4 °C. The immune complex was detected using Clean-Blot IP detection reagent (Pierce Biotechnology) and visualized using the Amersham ECL detection reagents (GE Healthcare, Chicago, IL, USA).
