The plasmids designed in this study are summarized in Supplementary Table 1. Mammalian vectors used in this study used plasmids designed by us earlier as templates16 (link). Anti-GFP nanobody vhhGFP4, referred in this paper as “anti-GFP iB” or “iB(GFP)” was from pcDNA3_NSlmb-vhhGFP4, a gift from M. Affolter (Addgene plasmid #35579). pQP-2376 was used as an intermediate cloning step, NSImb domain was further removed, to get pQP-2460. Vector containing PiggyBac52 (link) terminal repeats and EGFP-PAC under CMV promoter, as well as vector for hyperactive PiggyBac transposase expression were constructed by VectorBuilder (Cyagen Biosciences). pLentiEKAR2G2 was a gift from O. Pertz (Addgene plasmid #40178). pcDNA3-AktAR2 was a gift from J. Zhang (Addgene plasmid #64932). GFP-ERK2 was a gift from R. Seger (Addgene plasmid #37145). mEGFP-HRAS plasmid was a gift from K. Svoboda (Addgene plasmid #18662). Anti-RAS monobody NS1, referred in this paper as “iB(RAS)” was synthetized by GenScript, using amino acid sequence from 5E95 Protein Data Bank entry for reverse translation53 (link). Actin chromobody, referred in this paper as “iB(actin)” is commercially available from ChromoTek.
