Cck 8 labelling reagent

Manufactured by Dojindo Laboratories
Sourced in Japan

The CCK-8 labelling reagent is a colorimetric assay kit used to measure cell viability and proliferation. The reagent contains a water-soluble tetrazolium salt that can be reduced by dehydrogenase enzymes in viable cells, producing a colored formazan dye that can be quantified spectrophotometrically.

Quantifying Cisplatin's Cytotoxicity on HCC Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Briefly, HCC cell lines were seeded onto a 96-well plate (3 × 103 cells/well) 12 h prior to treatment with various concentrations (0–512 μg/ mL) of cisplatin (Sigma-Aldrich, St. Louis, MO, USA)29 . The cells were grown in a final 100-mL volume of culture medium per well. CCK-8 labelling reagent (10 μL; Dojindo Molecular Technologies Inc, Kumamoto, Japan) was added to each well and incubated for 1 h. Absorbance of the formazan product was measured at 450 nm. To achieve significance in quantitative analysis, experiments were repeated at least three times.

Cell Viability Assay of MDA-MB-231 and MCF-7

Check if the same lab product or an alternative is used in the 5 most similar protocols
A total of 2×103 MDA-MB-231 or MCF-7 cells in 100 µl high-glucose DMEM culture solution per well were added into a 96-well plate and incubated overnight. Subsequently, it was tested daily for ≤6 days. Prior to testing, 90 µl serum-free medium with 10 µl CCK-8 labelling reagent (Dojindo, Molecular Technologies, Inc.) was added into each well and incubated for 2 h in the dark at 37°C. The viability of the cells was then analysed by measuring their absorbance at 450 nm using an enzyme–labelled meter (Thermo Fisher Scientific, A33978, Inc.).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
