Metabolic Profiling and Insulin Sensitivity

Body weight was monitored every week post-surgery (week 0). On week three, animals were separated into individual cages to adapt to their surroundings for one day. Food intake was measured on the third day. During week 6, all the animals were placed into metabolic cages (Columbus Instruments, Comprehensive Lab Animal Monitoring System (CLAMS), OH, USA) to adapt to their surroundings 12 hours before measurement. All metabolic data was collected in night phase. For the insulin tolerance test during week 6, animals were fasted overnight. The baseline plasma glucose was measured by glucometer (FreeStyle Lite system, Abbott Diabetes Care Inc. CA, USA) at t=0. Then 0.75U/kg insulin was systemically injected immediately after baseline samples. The plasma glucose was measured at t=15, 30, 60, and 120 min after insulin injection.

ITT was conducted in mice after fasting of 6 hours as described elsewhere 45 (link). At Time 0, blood glucose was measured to set a baseline. insulin (1 unit/kg ITT) was injected i.p., blood samples were collected from the tail vein at 15, 30, and 60 minutes. The plasma glucose was measured by a glucometer (FreeStyle Lite system, Abbott Diabetes Care CA, USA).

Quantifying Metabolic Markers in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
For blood glucose measurement, a drop of tail blood was applied by glucometer (FreeStyle Lite system, Abbott Diabetes Care Inc., CA, USA). For insulin measurement, about 150 μl of trunk blood was collected before scarification. The plasma samples were harvested and froze until ELISA by centrifuging the blood samples (with anticoagulant) at 1500 rpm for 15 min (4°C). ELISA was performed following the procedure provided by ELISA kits (EMINS, Thermo Fisher Scientific).
For GLP-1 measurement, we express AAV-DIO-hM3Dq in the NTS of Gcg-Cre mice. After intraperitoneally injecting DREADD agonist CNO, we collected the fresh DMH brain slice (300 μm slice × 2) in lysis buffer and then performed ELISA (CSB-E08118, CUSABIO) according to the provided procedure with kits.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
