Quantitative UGT Enzyme Assay Protocol

3'-Phosphoadenosine-5'-phosphosulfate (PAPS) was purchased from R&D Systems. Rat liver microsomes and cytosols were acquired from Gibco CellCite. NADPH solution A, NADPH solution B, UGT Reaction Mix solution A, and UGT Reaction Mix solution B were purchased from Corning Gentest. Tris-HCl buffer (KD Medical) and PBS buffer (Gibco) were used as purchased. Polyethylene glycol 400 was from Sigma-Aldrich Corporation. Ethylenediaminetetraacetic Acid (EDTA) was purchased from Acros Organics through Fisher Scientific. Trans-tamoxifen-13C2, 15N was purchased from ISOTEC through Sigma-Aldrich Corporation; Fulvestrant was from Cayman Chemical Company; ZB716 was synthesized in our laboratory with over 99% purity [27 (link)]. Water (HPLC grade), acetonitrile (HPLC grade), methanol, formic acid, dimethyl sulfoxide (DMSO), and other chemicals were from Fisher Scientific.

Cell Culture Conditions for A549 and H1793 Cell Lines

A549 and H1793 cells were purchased from the American Type Culture Collection (Manassas, VA, USA) and grown for 2 weeks (five passages). A549 cells were cultured in 1640 medium supplemented with 10% fetal bovine serum (FBS) at 37°C with 5% CO2. H1793 cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM)/F12 medium supplemented with 5% FBS, 0.005 mg/ml insulin, 0.01 mg/ml transferrin, 30 nM sodium selenite, 10 nM hydrocortisone, and extra 2 mM l-glutamine at 37°C with 5% CO2. For experiments, cells were plated in phenol red-free medium that was supplemented with either charcoal-stripped serum or no serum for at least 24 h prior to adding the ligand, and 48 h later they were treated with the following agents: E2 (Sigma-Aldrich, St. Louis, MO, USA), (1-(4-(6-bromobenzo[1,3]dioxol-5-yl)-3a,4,5,9b-tetrahydro-3H-cyclopenta[c]quinolin-8-yl)-ethanone, 1-[(3aS,4R,9bR-rel)-4-(6-bromo-1,3-benzodioxol-5-yl)-3a,4,5,9b-tetrahydro-3H-cyclopenta[c]quinolin-8-yl]-ethanone) (G1, a GPER agonist; Cayman Chemical, Ann Arbor, MI, USA), fulvestrant (an ER antagonist and also a GPER agonist; Cayman Chemical), and (4-(6-bromo-benzo[1,3]dioxol-5-yl)-3a,4,5,9b-tetrahydro-3H-cyclopenta[c] quinoline) (G15, a GPER antagonist; Cayman Chemical) at the indicated concentrations. For cell culture experiments, all reagents used were formulated in 100% dimethyl sulfoxide.

Breast Cancer Cell Lines and Culture

Tamoxifen-resistant (MCF7-TamR) was purchased from Merck (Dietikon, Switzerland), fulvestrant-resistant (MCF7-TamR) was purchased from AXOL (Cambridge, United Kingdom), parental ER+ breast cell lines [MCF7-Control (Ctrl)] and T47D were purchased from the American Type Culture Collection (ATCC, Manassas, VA, USA). T47D was treated with Tamoxifen (Sigma, St. Louis, MO, USA) or fulvestrant (Cayman Chemical, East Ellsworth Rd., MI, USA) and selected resistance cells, T47D-TamR and T47D-FulR, respectively. MCF7-TamR and MCF7-FulR cells were cultured in DEME/F12 medium with low glucose and 1% of FBS containing 1 μM of Tamoxifen or 0.1 μM fulvestrant. MCF7 and T47D cells were cultured in DMEM (Dulbecco's modified Minimal Essential Medium) medium with high glucose and 10% of FBS. T47D-TamR and T47D-FulR were cultured in DMEM with high glucose and 10% of FBS containing 1 μM of Tamoxifen or 0.1 μM of fulvestrant. The human normal breast cell line, H184B5F5/M10 was obtained from the Bioresource Collection and Research Center (Hsinchu, R.O.C). H184 cells were cultured in MEM-alpha medium with 10% FBS.

Pharmacological Evaluation of Gastrointestinal Strips

Charybdotoxin was purchased from Tocris Bioscience (Minneapolis, MN, USA). All agents were dissolved in either distilled water or dimethyl sulfoxide (DMSO). The amount of DMSO or distilled water (10 µL) injected to the chambers had no effect on the strips [12 (link)].

Estrogen, Tmx, and fulvestrant effects

Estrogen, tamoxifen (Tmx) and fulvestrant were dissolved in 100% DMSO at a concentration of 1 mM. Forty-eight hours after isolation, in separate experiments, CF were either treated with 1 µM E2, (Cayman Chemical, MI, USA) or 250 µM Tmx (Cayman Chemical, MI, USA) or 100 nM fulvestrant (Sigma Aldrich, ON, Canada) for 30 min before incubating with Rsv for 24 h. The live and dead assay was performed after the 24 h of treatment as described earlier.

Estradiol Dose Optimization in Cell Culture

Cultured cells were treated with varying doses of estradiol (E2) (10,006,315, Cayman Chemicals) to determine the optimal working concentration. The cells were seeded in 6-well plates, and cultured in media supplemented with either 20, 10, 1, 0.1, 0.001 μM E2 for 24 h. Dimethylsulfoxide (DMSO) and tamoxifen (4-hydroxytamoxifen) were purchased from Sigma Chemical Co. (USA). Fulvestrant was purchased from Cayman Chemical (USA). Cells were treated at an indicated concentration of the antagonists and cell lysates analyzed by immunoblotting.
