Pharmaceutical Composition Analysis

TFV and FTC drug substances were obtained from Gilead Sciences (Foster City, CA, USA). Tablet excipients (mannitol, microcrystalline cellulose, crospovidone, hydroxyethyl cellulose, and sodium stearyl fumarate) were either United States Pharmacopeia (USP) or National Formulary (NF) grade. TFV-d6, cladribine, and emtricitabine-13C15N2, and (−)-emtricitabine-13C15N213CH10FN15N2O3S (internal standards) were obtained from Toronto Research Chemicals, Inc. (Toronto, ON, Canada). TFV–diphosphate (TFV-DP) and TFV-DP, adenine-13C5 were purchased from Moravek Biochemicals, Inc. (Brea, CA, USA). Weck-Cel® surgical spears were obtained from Medtronic Ophthalmics (Jacksonville, FL, USA). Organotypic human vaginal-ectocervical tissues (EpiVaginal™ tissues; VEC-100-FT) were obtained from MatTek Corporation (Ashland, MA, USA). Dulbucco’s phosphate buffered saline (DPBS) was obtained from Invitrogen (Carlsbad, CA, USA). Gynol II (Advanced Care Products, Ortho Pharmaceutical Corp., Raritan, NJ, USA) was purchased commercially.

TAF and TFV Characterization Protocol

TAF (also known as GS-7340) was provided by Gilead Sciences, Inc. TAF and TFV and their stable isotopically labelled (SIL) internal standards (TFV-d6 and TAF-d5) were obtained from Toronto Research Chemicals (Canada). Poly(vinylpyrrolidone) (PVP K29-32; and PVP K90) was provided by Ashland (Kidderminster, UK). Poly(vinyl alcohol) (PVA 9–10 ​kDa) and solvents methanol ≥99.9%, dimethyl sulfoxide (DMSO), tetrahydrofuran (THF) inhibitor-free ≥ 99.9% and acetonitrile (ACN) ​≥ ​99.9% (all HPLC grade), ammonium acetate ≥98%, and Acetic acid ≥99.7% were purchased from Sigma-Aldrich, St. Louis, USA. Two different types of poly(d,l-lactide-co-glycolide) (PLGA) were used, a low viscosity PLGA, Purasorb® PDLG 7502 with a lactide:glycolide ratio of 75:25 and ester end (Purac®, Corbion, Netherlands) (LV-PLGA) and a high viscosity PLGA with a lactide:glycolide ratio of 75:25 and ester end (Viatel® DLG 7503 ​E) (HV-PLGA). Both PLGAs were a donation from Ashland (Kidderminster, UK). All other reagents used in this work were of analytical grade and purchased from Sigma-Aldrich. Elga purified water was used in all cases (Purelab option® Elga LabWater, High Wycombe, UK).

Synthesis and Characterization of Tenofovir Alafenamide Polymer Conjugates

Materials were purchased from Sigma-Aldrich unless otherwise specified. All solvents were Fisher HPLC grade. Chain transfer agents, 4-(((2-carboxyethyl)thiocarbonothioyl)thio)-4cyanopentanoic acid (CCC) and 4-cyano-4-((ethylsufanylthiocarbonyl)-sulfanyl)pentanoic acid (ECT) were purchased from Boron Molecular and OMM Scientific (Texas, USA), respectively.
Glycerol monomethacrylate was purchased from Polysciences and purified using basic alumina before polymerization. 2,2'-Azobis(4-methoxy-2,4-dimethylvaleronitrile) (V-70) was purchased from FUJIFILM Wako Pure Chemical Corporation, USA and used without further purification.
Fasudil hydrochloride was purchased from eNovation Chemicals (Green Brook, NJ, USA). Spectra/Por regenerated cellulose dialysis membranes were purchased from Spectrum Laboratories (Houston, TX, USA). Rhodamine B methacrylate (RhMA) was synthesized as described previously. [43] (link) Tenofovir alafenamide (TAF) fumarate (1:1 salt) was obtained from MedKoo Biosciences, Inc. Analytical standards: tenofovir (TFV) and tenofovir diphosphate (TFV-DP) were obtained from Cayman Chemical and Santa Cruz Biotech, respectively. Isotopic internal standards: TAF-d5 fumarate, TFV-d6 and TFV-DP 13 C5 were obtained from Toronto Research Chemicals and Moravek, respectively. HPLC grade methanol and water were purchased from VWR international Ltd.
