Isolation and Identification of Cariogenic Bacteria

The plaque samples were collected using the tips of sterile wooden toothpicks from carious lesions. The toothpicks were cut off and instantly dipped into 1 ml sterile phosphate-buffered saline (HiMedia, India) and stored at 4°C. Plaque samples were vortexed for a minute to disperse the plaque and obtain a homogeneous suspension. The samples were diluted by 100-fold in 1x sterile phosphate-buffered saline and plated on Mitis Salivarius bacitracin (MSB) agar. The MSB agar composed of mitis salivarius agar (HiMedia, India) and supplemented with 15% of sucrose, 1% of agar, 0.0001% potassium tellurite solution and 0.2 units/ml of bacitracin (HiMedia, India). The plates were incubated anaerobically at 37°C for 48 h.[19 (link)]
After the incubation period, the colonies were identified on the basis of colony morphology.[20 (link)] The typical colonies from each sample plate were transferred to brain–heart infusion (BHI) broth (HiMedia, India) and incubated at 37°C for 18 h. After the incubation period, the broth cultures were streaked on MSB agar and anaerobically incubated at 37°C for 48 h. The overnight bacterial cultures were stored in 80% glycerol stock at −20°C.

Antimicrobial Resistance Profiling of MRSA

Resistance to antibiotics was confirmed as per the guidelines of internationally recognized standards of the Clinical and Laboratory Standards Institute (CLSI)53 . MRSA, MRSE and MRSH strains were screened for susceptibility by 22 antimicrobial agents using disc diffusion method on Muller-Hinton agar. The antibiotics disc tested were (concentration in μg, HiMedia, India) Erythromycin (E-15 μg), Clindamycin (CD-2 μg), Gentamicin (GEN-10 μg), Cefotaxime (CTX-30 μg), Norfloxacin (NX-10 μg), Ciprofloxacin (CIP-5 μg), Tetracycline (TE-30 μg), Rifampicin (RIF-5 μg), Nitrofurantoin (NIT-300 μg), Linezolid (LZ-30 μg), Chloramphenicol (C-30 μg), Amoxyclav (AMC-10 μg), Amphicillin (AMP-10 μg), Azithromycin (AZM-15 μg), Bacitracin (B-10 Units), Cefoxitime (CFM-5 μg), co-Trimoxazole (COT-23.75 μg), Kanamycin (K- 30 μg), Oxacillin (OX-I μg), Penicillin G (P-10 Units), Streptomycin (S-100 μg), Tetracycline (TE-30 μg) and Tobramycin (TOB-30 μg). The results were recorded after 24 h of incubation at 37 °C and interpreted as per CLSI guidelines. Screened isolates showing resistance to more than three classes of non-β-lactam antibiotics were considered as multidrug resistant (MDR) strains, as described by Chajecka et al.,9 (link). Assay was performed in triplicates for all strains.

Anaerobic Media Composition for Microbial Cultures

The composition of media per 500 ml was as follows: Columbia agar base (21.25%) (code LAB001, UK), 5 µg/mL of hemin (1.5 mg) (Sigma-Aldrich, China), 1 µg/mL vitamin K1 (0.5 mg) (Himedia), 5% human blood (25 ml), colistin methanesulfonate (7.68 mg), bacitracin (5 mg) (Himedia), nalidixic acid (7.5 mg) (Himedia), and distilled water [24 (link)].
Media were prepared and stored under strictly anaerobic conditions to prevent oxidation.

Brucella Detection in Dairy Cattle Milk

Isolation of Chronic Periodontitis P. gingivalis

This study involves clinical strain of P. gingivalis Gram-negative bacteria isolated from periodontal pocket of patients attended Shorsh dental center in Sulaymaniya city/Iraq in August 2017, subgingival plaque samples were collected from systematically healthy persons suffering from chronic periodontitis, from periodontal pockets with of at least six mm depth with sterilized paper point, then the samples were spread on Colombia agar media (LAB001 UK) supplemented with sheep blood %5, 5μg/ml hemin (sigma Aldrich-china), 1μg/ml vitamin K1 (Himedia), 7.68 mg colistin methanesulfonate (Himedia), 5 mg bacitracin (Himedia), and 7.5 mg nalidixic acid (Himedia) according to [18 ]. Plates were incubated for 7-10 days anaerobically using an anaerobic jar and anaerobic gas packs (AnaeroGen® system Oxoid), patients' consent and approvals were obtained prior to collecting the samples.

Streptococci Identification Media Preparation

The mitis salivarius agar powder recommended for Streptococci and bacitracin were obtained from HiMedia Laboratories Limited, Mumbai. The process of making culture media was carried out as per the instructions provided by the manufacturer while addition of bacitracin was done as recommended by Gold et al., 1973.[9 (link)] One ml of sterile 1% potassium tellurite solution was also added to it for staining the colonies. The pH of the agar was maintained at 7.4 at 25°C.

Antibiotic Susceptibility of Probiotic Isolates

The following antibiotics were tested in the form of discs: bacitracin (10 µg) (HiMedia), cefpirome (30 µg) (Oxoid), chloramphenicol (30 µg) (Oxoid), clarithromycin (15 µg) (Oxoid), penicillin (10 µg) (Oxoid), sulfamethoxazole/trimethoprim (STX) (25 µg) (Oxoid), tetracycline (30 µg) (Oxoid), and vancomycin (30 µg) (Oxoid). The plates were incubated at 30 °C for 24 h under limited oxygen and aerobic conditions for LAB and yeast, respectively, and diameter of the inhibition zones was measured by using vernier caliper.
