Gamma Scintigraphy for Sustained Release Depot Evaluation

Gamma scintigraphy was carried out to confirm the development of sustained release depot at the site of injection following the administration of liquid ZP-LPS formulation. Briefly, ZP was radiolabeled with 99mTechnetium-pertechnetate (Tc-99M) by an in-house developed standard procedure using stannous chloride as reducing agent. Gamma Scintigraphy Confirms Intranasal CONPs Brain Targeting

The gamma scintigraphy (Figure 3) was used to confirm the brain targeting of intranasal CONPs. Considering proof-of-concept study, only three healthy male Sprague Dawley rats of weight approx. 250 ± 30 g, aged 6–7 months were used. Twenty four hours before of the experiment, rats remained in a fasted state. Further, radiolabeled intranasal CONPs (50 µCi of 99mTc) were administered to rats using micro pipette. To understand the brain delivery of CONPs, the images were obtained at 15, 30 and 60 min using gamma camera (Siemens T2 SPECT-CT).
