Analytical Methods for Glucosinolate Analysis

Reagents were of analytical purity. Media components (peptone, glucose monohydrate, sodium nitrate, potassium hydrogenphosphate, magnesium sulfate, potassium chloride, ferrous sulfate) were from Reanal (Budapest, Hungary). Allyl isothiocyanate, allyl cyanide, 2-phenylethyl isothiocyanate and phenylpropionitrile and glutathione (reduced form) were from Sigma Aldrich (MO, USA). Pure glucosinolates were from Phytoplan (Germany). LC-MS grade acetonitrile, water and formic acid were purchased from Fisher Scientific (Belgium). Solvents (1-propanol, 2-propanol) and media additives (streptomycin, chloramphenicol and dichloran) were from VWR. Double distilled water was used throughout the study.

(R)-5-Vinyloxazolidine-2-thione (R-OZT) was purchased from Biosynth AG (Staad, Switzerland). 4-Hydroxybenzyl GLS (≥99%) and methylene chloride (GC Ultra grade) was obtained from Carl Roth GmbH (Karlsruhe, Germany). Acetonitrile (Ultra Gradient HPLC grade) was procured from J.T. Baker (Deventer, The Netherlands). NaSO4 (≥99%) and methanol (>99.9) was purchased from VWR International GmbH (Darmstadt, Germany).
All solvents were of LC-MS or GC-MS grade, water was of Milli-Q quality.

Extraction and Characterization of Nematode Juveniles

Meloidogyne hapla was maintained on tomato (Solanum lycopersicum) cv. Moneymaker under greenhouse conditions (25/19 °C, 60% humidity, 15/9 h day night cycle). Freshly hatched second-stage juveniles (J2) were extracted from heavily galled root systems placed under a mist chamber (22 °C) [46 ]. Hatched J2 were collected every day and stored at 6 °C until being used in experiments, but no longer than 10 days.
The ITCs allyl isothiocyanate, benzyl isothiocyanate, butyl isothiocyanate, ethyl isothiocyanate, methyl isothiocyanate, phenyl isothiocyanate, and 2-phenylethyl isothiocyanate were obtained from Sigma-Aldrich (St. Louis, MO, USA) and diluted with sterilized water to achieve the concentrations requested in the different experiments. For the allyl isothiocyanate (AITC) experiment in soil, the volumetric measure of the pots was used to calculate the concentration (µmol/mL) applied to the soil.

Type I (18.2 MΩ cm−1) water purified by a Human Zeneer Power I water purification system (Human corporation, Seoul, Korea) was used throughout the study.

Behavioral Bioassays of Volatile Compounds

Garlic oil, A. sativum, was purchased from Elixarome Ltd; rosemary, R. officinalis, and lavender, La. angustifolia, essential oils were provided by Botanix Ltd, and bog myrtle oil, Myrica gale, was purchased from Amphora Aromatics Ltd. Diallyl sulphide, diallyl disulphide, allyl, propyl, butyl and 2-phenylethyl isothiocyanate were purchased from Sigma-Aldrich. 4-Pentenyl isothiocyanate was synthesized from 4-pentenylamine hydrochloride as described previously (Dawson et al. 1993) . Solutions of extracts and individual compounds in absolute ethanol were prepared (0.1, 0.05, 0.01 and 0.001 mg mL 1 ) and diluted to 1 lL L 1 in ASW (Ingvarsdottir et al. 2002b) to give a final concentration of 100, 50, 10 and 1 parts per trillion, respectively, for use in Y-tube behavioural bioassays.
