Establishing Ovarian Cancer Cell Lines

Human EOC cell lines (OVCAR3, SKOV3, and A2780) were obtained from Shanghai Genechem Co., Ltd. (Shanghai, China). The normal human ovarian epithelial cell IOSE80 was bought from Yuchicell (Shanghai) Biological Technology Co., Ltd. (Shanghai, China). Cells were cultivated in RPMI 1640 (HyClone Laboratories, Logan, UT, USA) medium, supplemented with 10% fetal bovine serum (FBS) (Gibco, Grand Island, NY, USA), 2 mM L-glutamine, 100 U/ml penicillin, and 100 U/ml streptomycin (all from (Biological Industries, Beit HaEmek, Israel) at 37°C and 5% CO2. All the stable transgenic cell lines were maintained below five passages.

Establishing DEPDC1B Overexpression Cell Lines

The human normal ovarian cell line IOSE-80 was obtained from Shanghai Jingyuan Co., Ltd. (Shanghai, China). The EOC cell lines OVCAR3 and SKOV3, and the human hepatocellular carcinoma cell line BEL-7404 were obtained from Shanghai GeneChem Co., Ltd. (Shanghai, China). The human lung carcinoma cell line A549 was purchased from Cobioer Biosciences Company (Nanjing, China). IOSE-80 and BEL-7404 cells were routinely maintained in RPMI 1640 medium (GIBCO, Grand Island, NY, USA) supplemented with 10% fetal bovine serum (FBS, GIBCO). OVCAR3 cells were maintained in RPMI 1640 medium supplemented with 20% FBS. SKOV3 cells were maintained in McCoy's 5a medium (GIBCO) supplemented with 10% FBS. A549 cells were incubated in F12K medium supplemented with 10% FBS. Lentiviral DEPDC1B overexpression vector was purchased from the GeneChem. To establish cell lines stably expressing a high level of DEPDC1B, lentivirus-infected cells were selected with 2 μg/mL of puromycin (GeneChem).

Olaparib Sensitivity in Ovarian Cell Lines

A2780 and A2780cisR (cisplatin resistant) human ovarian cell lines were gifts from Fudan University Shanghai Cancer Center. IOSE‐80 and HEY‐T30 ovarian cell lines originated from a gift from Dr Luopei Guo (Obstetrics and Gynecology Hospital of Fudan University). ES‐2, SKOV3, OVCAR3, and CAOV3 ovarian cell lines were purchased from GeneChem (Shanghai, China). All cell lines were cultured according to ATCC guidelines at 37°C in a 5% CO2 incubator. The olaparib (Selleck, catalog number S1060) was dissolved in DMSO, and the final concentration of DMSO in the medium was 0.1%. After the cells were plated for 24 h, cells were overlaid with (0, 2, 10, 25, 50 μM) olaparib‐conditioned medium and harvested for 24 h.
