Peptide Synthesis and Digestion Assay

The peptide AVFQHNCQE was synthesized by CASLO Aps. (Kongens Lyngby, Denmark) and its purity was 98.94%. Captopril was provided by Santa Cruz Biotechnology (Dallas, TX, United States). Pepsin, bile salts mixture, trypsin, α-chymotrypsin, porcine pancreatic lipase, colipase, trifluoroacetic acid (TFA), formic acid (FA) LC-MS grade, and bovine serum albumin (BSA) were obtained from Sigma-Aldrich (Madrid, Spain). Acetonitrile and methanol LC-MS grade were purchased from Merck (Darmstadt, Germany). The synthetic isotopically labeled peptide IFV*TGQDYNDK (V* = Val-13C5,15N) was obtained from Biomatik (Ontario, Canada) and used as internal standard (IS) for MS experiments. Chicken foot hydrolysate Hpp11 was obtained by our group following the procedure described by Bravo et al. [22 ,24 (link)]. All other chemical solvents used were of analytical grade.

Enzymatic Hydrolysis and Antioxidant Evaluation

Flavourzyme® 1000 L (EC 3.4.11.1, 500 leucine amino peptidase units (LAPU)/g from Aspergillus oryzae) was kindly provided by Novozymes (Bagsværd, Denmark). ACE (EC 3.4.15.1), 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH), picrylsulfonic acid and acetonitrile for HPLC were purchased from Sigma-Aldrich (Madrid, Spain). Captopril (PubChem CID: 44093) and resveratrol were provided by Santa Cruz Biotechnology (Dallas, TX, USA) and Carl Roth (Karlsruhe, Germany), respectively. Folin–Ciocalteu reagent, coumaric acid, quercetin, gallic acid, catechin, and epicatechin were purchased from Fluka/Sigma-Aldrich. 4-Hydroxybenzoic acid, caffeic acid, procyanidin dimer B2, vanillic acid, and malvidin-3-O-glucoside were purchased from Extrasynthése (Lyon, France). Ferulic acid, cyanidin-3-O-rutinoside, and peonidin-3-O-rutinoside were purchased from PhytoLab (Vestenbergsgreuth, Germany). Analytical grade reagents were always used.

Transaortic Constriction and Pharmacological Interventions in Mice

All experimental procedures were designed in accordance with the National Institutes of Health guidelines and approved by the Animal Welfare Committee and the Center for Laboratory Animal Medicine and Care in the University of Texas Health Science Center at Houston. Ten to eleven-week-old male C57BL/6J wild-type mice were purchased from Jackson Laboratory, Bar Harbor, ME, USA. At the age of 12 weeks, mice weighing 23-31grams (27.5±1.9g) were randomized to treatment with or without various drugs in the drinking water for three days, then underwent TAC or sham operation. Treatment was continued for two weeks post-operation. Losartan (sc-204796A, Santa Cruz Biotechnology, 0.6g/L)12 (link), captopril (sc-200566A, Santa Cruz Biotechnology, 75mg/L)11 , AT2R agonist C21 (a gift from Vicore Pharma, 300μg/kg/day),25 (link) AT2R antagonist PD123319 (P186-10MG, Sigma-Aldrich, 3mg/kg/day)20 (link) and MasR agonist AVE0991 (a gift from Sanofi-Aventis, Frankfurt/Main, 576μg/kg/day)26 (link) were administrated 1 hour prior operation to two weeks post-operation via intraperitoneal injection. Chicken feet from Gallus gallus domesticus were provided by a local farm (Granja Gaià, La Riera de Gaià, Spain). Protamex® (Novozymes, Bagsværd, Denmark) (EC 3.4.21.62 and 3.4.24.28, 1.5 AU/g from Bacillus licheniformis and Bacillus amyloliquefaciens), was kindly provided by Novozymes (Bagsværd, Denmark). ACE (angiotensin-converting enzyme, EC 3.4.15.1), N-Hippuryl-His-Leu (Hip-His-Leu), Captopril (PubChem CID: 44093) were purchased from Santa Cruz Biotechnology (Dallas, TX, USA) and o-aminobenzoylglicil-p-nitrofenilalanilprolina (o-Abz-Gly-p-Phe(NO2)-Pro-OH, PubChem CID: 128860) was provided by Bachem Feinchemikalien (Bubendorf, Switzerland). Acetylcholine (PubChem CID: 187), methoxamine hydrochloride (PubChem CID: 6081) and heparin heparin (PubChem CID: 772) were purchased from Sigma-Aldrich (Madrid, Spain). All other chemical solvents used were of analytical grade.

Functionalized PEG-Ang Peptide Conjugation

Hoechst 33258 was obtained from Polysciences Inc. (Warrington, PA, USA). Integrin‐α8 goat antibody was purchased from R&D Systems (Minneapolis, MN, USA). Cy2‐anti‐goat secondary antibody was obtained from Jackson Immuno Research Labs (PA, USA). CXN2‐HA‐AT1R‐YFP was a gift from Yusuke Ohba (Addgene plasmid #101659; http://n2t.net/addgene:101659; RRID: Addgene_101659). All other materials and reagents in analytical grade were obtained from Sigma‐Aldrich (Taufkirchen, Germany).
