Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) Protocol

Human serum, bovine serum albumin (BSA), N-alpha-tosyl-L-lysinyl-chloromethylketone (TLCK) and Lucifer yellow (LY), Digoxin, Prazosin, Atenolol and Atenolol-d7 were from Sigma Aldrich (Saint-Quentin Fallavier, France). Collagenase/dispase and DNAse I were from Roche Applied Science (Basel, Switzerland). Digoxien-d3 was from Artmolecule (Poitier, France).
All chemicals used for ICP-MS were of analytical grade. Nitric acid was used to prepare 0.2 % HNO3 (v/v) with ultrapure water. All single element stock solutions (1000 mg/L) were delivered by SCP Science and certified for purity and concentration. From these stock solutions, a mixed working standard solution with a concentration of 10 mg/L for each element was prepared by putting 1 mL of each stock solution in a 100 mL measuring flask, adding 5 mL of purified HNO3 and diluting to 100 mL with ultrapure water (MilliQ, Millipore, Germany).

Comprehensive Pharmaceutical Analysis Protocol

The 11 pharmaceuticals studied are classified following their Anatomical Therapeutic Chemical Classification code (ATC). They all were purchased from Sigma-Aldrich (St. Louis, MO, USA) with a purity range of 98–99%. Furthermore, atenolol-d7, lidocaine-diethyl-d10 and acetaminophen-methyl-d3 were acquired also at Sigma-Aldrich and were used as internal standards. HPLC grade methanol and acetonitrile (ACN) were supplied by VWR Chemicals (Leuven, Belgium). Ammonium formate (NH4COOH) and ammonium hydroxide (NH4OH) were supplied by Sigma-Aldrich (St. Louis, MO, USA). Hydrochloric acid 37% (HCl) and formic acid (HCOOH) were purchased from Fisher Scientific (Bridgewater, MA, USA). Ultra-pure Milli-Q water was obtained through a Millipore purification system (Millipore, Bedford, MA, USA). Standard stock solutions were prepared in amber glass vials at a concentration of 1000 mg L−1 in methanol, and working solutions were prepared between 1 and 2500 µg L−1 in 90% of Milli-Q water and 10% of methanol.

Atenolol Pharmacokinetic Analysis

S-(−)-atenolol and atenolol-D7 were purchased from Sigma-Aldrich (St. Louis, MO, USA). Isoflurane was obtained from Baxter Medical AB (Kista, Sweden). Ringer’s solution was prepared to perfuse microdialysis probes and comprised 145 mM NaCl, 0.6 mM KCl, 1.0 mM MgCl2, 1.2 mM CaCl2, and 0.2 mM ascorbic acid in 2 mM phosphate buffer (pH 7.4). Normal saline was obtained from Braun Medical AB (Stockholm, Sweden), and water was purified using a Milli-Q system (Millipore, Bedford, MA, USA). Ammonium acetate and acetonitrile were purchased from Merck (Darmstadt, Germany). All other chemicals were of analytical grade.

Pharmaceutical Target Compounds Analysis

Based on the marketing data of pharmaceuticals, 57 types of pharmaceuticals were initially selected as target compounds in our preliminary study. Out of the 57 compounds, we selected 27 compounds as the targets in this study based on their influent concentrations and detection frequencies. In total 27 types of pharmaceuticals, including 11 antibiotics, 6 anti-inflammatory/NSAIDs, 2 antiarrhythmic drugs, and 1 stimulant were selected as the target substances. Table 1 shows the physicochemical properties of the substances. For the standard substances and reagents used in the analysis, 10 mg/L of a standard solution of 98% or higher purity purchased from Sigma-Aldrich (Saint Louis, MO, USA) and Fluka (Seelze, Germany) was prepared and stored in a freezer at −20 °C or below. It was diluted and used on the day of the analysis. Twenty types of isotopically labeled standards (acetaminophen-d4, salicylic acid-d4, atenolol-d7, caffeine-13C3, carbamazepine-d10, cimetidine-d3, ciprofloxacin-d8, diclofenac-13C6, diphenhydramine-d3, erythromycin-d6, gemfibrozil-d6, ibuprofen-d3, iopromide-d3, ketoprofen-d3, naproxen-d3, ofloxacin-d3, propranolol-d7, sulfadimethoxine-13C6, testosterone-d3, and trimethoprim-d9) were purchased from Sigma-Aldrich and Toronto Research Chemicals (Toronto, ON, Canada).

Analytical Reagents for Toxicology Research
