(-)-epigallocatechin-3-gallate (EGCG), N-acetylcysteine (NAC), and N-vanillylnonanamide (NVN) were obtained from Sigma Aldrich Co. Ltd. (St. Louis, MO). Recombinant human IL-1β was purchased from GIBCO BRL (Grand Island, NY). The inhibitors, BAY 11–7085 for NF-κB, SB 203580 for p38, and PD 98059 for MEK 1, were purchased from Calbiochem (La Jolla, CA). EGCG and NAC were dissolved in H2O. NVN, BAY 11–7085, SB 203580, and PD 98059 were dissolved in dimethyl sulfoxide, methyl alcohol, or H2O. The final vehicle concentration was adjusted to 0.1% (v/v), and the control medium contained the same quantity of vehicle. The ginger capsules contain 6.55% 6G, 0.94% 8G, 1.88% 10G, 2.21% 6S, 0.37% 8S, and 0.95% 10S. β-Glucuronidase from bovine liver, sulfatase from Helix pomatia, d-saccharic acid 1,4-lactone monohydrate (β-glucuronidase inhibitor), ascorbic acid, and N-vanillylnonanamide (internal standard, IS) were purchased from Sigma-Aldrich Inc. (St Louis, MO, USA). Sodium acetate was purchased from Fisher Scientific. Nociceptive Pain Induction Techniques

Under 2% isoflurane anesthesia, a 30 G needle was inserted into the plantar surface of the paw subcutaneously. Fifty microlitres of BiTox (200 ng in 0.9% NaCl solution) or vehicle or 50 μL of CFA (Complete Freund's adjuvant, SIGMA, Gillingham, United Kingdom) were injected for more than 10 to 20 seconds. Fifty microlitres of 2% formalin or 10 μL of 0.3% capsaicin N-vanillylnonanamide (synthetic capsaicin; SIGMA) were injected without anesthesia. For the latter, the animal was lightly restrained in a glove during injection. Ten microlitres CFA was injected into the lateral ankle joint. For dose–response experiments, the concentrations of BiTox used were: 2, 50, 100, and 200 ng.
