Cytokine Profiling of Bone Marrow

For collecting bone marrow fluids, the four long bones (two femurs and two tibiae) of each mouse were flushed with the same 200 μl of PBS. Cells were spun down by centrifugation at 1,000 RPM for 5 min. Next, supernatants were clarified by a second centrifugation at 12,000 RPM for 5 min. IL-1α and IL-1β were measured by ELISA (mouse IL-1β and mouse IL-1α ELISA kit; R&D). IFN-α, IL-1β and other cytokines were measured using the Legendplex Anti-virus response panel (BioLegend).

Quantification of Cytokine Profiles

LPS-Induced Cytokine Response in Mice

Mice were intraperitoneally injected with LPS from Escherichia coli O55:B5 (L4005, Sigma-Aldrich). Blood was collected using EDTA-coated tubes at indicated time and then centrifuged at 1000g for 15 min at 4°C.
