used in experiments and HPLC analysis. Bis(p-nitrophenyl)
phosphate sodium salt, D-glucose-6-phosphate, glucose-6-phosphate
dehydrogenase, β-nicotinamide adenine dinucleotide phosphate
(NADP+), magnesium chloride hexahydrate, quinidine, sulfaphenazole,
furafylline, p-nitrophenol, ketoconazole, bicinchoninic
acid (BCA) protein assay kit, and salts used for buffer preparation
were purchased from Sigma-Aldrich (St. Louis, MO).

Characterization of Kurarinone Metabolism

(Shanghai, China). Pooled liver microsomes from humans (HLMs), monkeys (MLMs), rabbits (RAMs), rats (RLMs), mouse (MOMs), dogs (DLMs), and minipigs (PLMs) were provided by the Research Institute for Liver Disease Co., Ltd. (Shanghai, China). Recombinant human supersomes (UGT1A1, UGT1A3, UGT1A6, UGT1A7, UGT1A8, UGT1A9, UGT1A10, UGT2B4, UGT2B7, UGT2B15, CYP1A2, CYP2C9, CYP2D6, CYP2E1, CYP3A4, and CYP2C19) were obtained from BD Gentest Corp. (Woburn, MA, USA).

Characterization of Cysteine Protease Inhibitors

Hemin chloride, chloroquine, quinacrine, amodiaquine, mefloquine, 8-hydroxyquinoline, quinine, quinidine and the β-carbolines norharman, tryptoline, harman, and harmine were obtained from Sigma-Aldrich. Indazole compounds 1,1´-[2,2´-biphenyldiyl)bismethylene]bis(5-nitro-1H-indazol-3-ol) (DIM-32) and 1,1´-(o-xylylene)bis(5-nitro-1H-indazol-3-ol) (DIM-5) were previously synthetized45 (link). 3,3′,5,5′-Tetramethylbenzidine (TMB), 2,2′-azinobis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) and tween 20 were obtained from Sigma and cysteine from Merck. DMSO and hydrogen peroxide (H2O2) were from Scharlau and Panreac, respectively. Globin was obtained from bovine hemoglobin (Sigma) by precipitation in acetone-0.1% HCl at low temperature89 (link),90 (link), and lab-stored crystallized and lyophilized bovine serum albumin (BSA) was from Sigma. cysteine proteases: papain from Carica papaya, ficin from fig tree latex and cathepsin B from bovine spleen were obtained from Sigma. The peptide Z-Phe-Arg-AMC was purchased from Bachem, and 7-amino-4-methylcoumarin (AMC) from Sigma.

Lysionotin and Testosterone Characterization

Coumarin (≥98%), diclofenac (≥98%), dextromethorphan (≥98%), and ketoconazole (≥98%) were purchased from ICN Biomedicals. Pooled HLMs were purchased from BD Biosciences Discovery Labware. All other reagents and solvents were of analytical reagent grade.

Luminescent CYP3A4 and CYP2D6 P450 Assays

The luminescent CYP3A4 P450-Glo™ and CYP2D6 P450-Glo™ assays and protocols were provided by Promega (Madison, WI, USA) (Cali et al. 2006 (link)). The reference drugs ketoconazole (KE) and quinidine (QD) were obtained from Sigma-Aldrich (St. Louis, USA). The enzymatic reactions were performed in white polystyrene, flat-bottom Nunc™ MicroWell™ 96-Well Microplates (Thermo Scientific, Waltham, MA USA). The luminescence signal was measured with a microplate reader in luminescence mode (EnSpire, PerkinElmer, Waltham, MA USA). The IC50 values of KE and QD were determined and calculated as we described previously (Sadek et al. 2016 (link)). The final concentrations of EMD 386088 were similar for both CYP3A4 and CYP2D6 assays from 0.025 to 25 μM.
