Quantification of Metabolites by ESI-MS

Both methanol and acetic acid were HLPC grade and bought from ROE Scientfic Inc. (Newark, U.S.A). All amino acids authentic compounds such as valine, histidine, arginine and lysine were purchased from Sinopharm Chemical Reagent Co. Ltd. (Shanghai, China). Creatine, L-carnitine (BR) and vitamin B6 (BR) were purchased from Shanghai Source Leaf Biological Technlogy Co., Ltd (Shanghai, China) with 98%, 98% and 99% purity, respectively. The deionized water used for experiments was provided by a Milli-Q water purification system (Billerica, USA). All standard solutions were prepared in deionized water and then used for ESI-MS analysis without further treatment.

Carboxylatopillar[5]arene Nanomaterial Synthesis

(R6G), citric acid, ethylenediamine, N-hydroxysuccinimide
(NHS), and 1-ethyl-3-(3-(dimethylamino)propyl)carbodimide hydrochloride

of 200 kV. Fourier transform infrared (FTIR) spectra were obtained
by using a Bruker Vertex 80 V spectrometer. Powder X-ray diffraction
(PXRD) measurements were conducted by using a PANalytical B.V. Empyrean
powder diffractometer. Ultraviolet–visible (UV–vis)
spectra were recorded using a Shimadzu UV-2550 instrument. Thermogravimetric
analysis (TGA) was performed on a Q500 instrument. Fluorescence spectra
were recorded in quartz cuvettes by using a Shimadzu RF-5301PC spectrophotometer.
ζ potentials were recorded on a Particle Sizing Systems Z3000
instrument.

HAMPT Combination Therapy Protocol

Stock solution of HAMPT is composed of the following: aspirin 75 mg (Aladdin Reagent Company, China), lysine 200 mg (Sinopharm Chemical Reagent Co., China), mifepristone 10 mg (Shanghai New Hualian Co., China), and doxycycline hyclate 50 mg (Shanghai Civi Chemical Technology Co., China). Drug doses were chosen based on the human doses of the four drugs [36 (link), 51 , 53 (link)-54 (link)] and converted to mouse doses according to the body surface area index [24 (link)]. The concentrations of the four drugs used in the in vitro assay were chosen based on the same dose ratio of the four drugs composed and used in the animal studies. The combination was so formulated to target different pathways of cancer metastasis.

Fabrication of PEN-PVP Nanofiltration Membranes

PEN ([η] = 1.15 dL g−1) synthesized in the laboratory as described in previous literature50 (link) was used as the base polymer in the casting solution. Reagent grade PVP-k30 supplied by Macklin Biochemical Technology Co. Ltd. was used as pore-forming agent. The chemical structure of PEN and PVP were shown in Fig. 1. Reagent grade DMAc (≥99.8% purity) and NMP (≥99.5% purity) supplied by Macklin, were used as mixed solvent without further purification. SiO2 NPs (7–40 nm, hydrophilic-380 type) and 3-glycidyloxypropyl (dimethoxy) methylsilane (GPMS, KH560) were obtained from Aladdin Biochemical Technology Co. Ltd. Lysine, Hydrochloric acid, Ethanol were purchased from Sinopharm Group Chemical Reagent Co. Ltd. Bovine serum albumin (BSA) with molecular weight of 68 000 Da was obtained from Energy Chemical Co. Ltd. Deionized water purified with Millipore system (Millipore, France) was used as the coagulation bath.
